摘　　要：目的研究C/EBPδ在结直肠癌中的表达情况及其对结直肠癌细胞增殖能力的影响和相关机制。方法选取2017年至2018年于云南省第二人民医院行结肠癌根治术的40例患者的肠癌组织以及对应的癌旁组织和正常肠组织,采用免疫组织化学法检测组织中的C/EBPδ蛋白质表达情况。采用免疫共沉淀方法检测结直肠癌组织中C/EBPδ与Siah2的相互作用。构建C/EBPδ过表达及干扰的病毒载体,分别感染结直肠癌Caco-2细胞,通过CCK8、流式细胞仪检测、平板克隆形成实验观察C/EBPδ过表达和干扰对细胞增殖、凋亡及周期的影响。采用卡方检验对阳性表达率相关因素进行统计学分析。结果(1)从正常肠组织到癌旁组织再到结直肠癌组织中C/EBPδ的表达依次升高,差异均有统计学意义(P<0.05),C/EBPδ蛋白表达量在伴有肝转移的结直肠癌患者中更为显著;(2)C/EBPδ升高的结直肠癌组织中伴有Saih2的升高,但二者没有直接的相互作用;(3)C/EBPδ过表达能显著提高Caco-2细胞的增殖,而C/EBPδ的shRNA干扰使结直肠癌细胞增殖明显减少;(4)与对照组相比,C/EBPδ的shRNA干扰显著抑制了结直肠癌细胞Caco-2细胞的体外迁移和侵袭能力(P<0.05)。结论C/EBPδ在结直肠癌组织中存在异常,过表达的C/EBPδ通过与Saih2相互作用促进结直肠癌细胞的侵袭和转转,其与结直肠癌的不良预后可能相关。Objective To investigate the expression of C/EBPδin colorectal cancer and its effect on the proliferation of colorectal cancer cells.Methods The expression of C/EBPδprotein in 40 patients undergoing the radical resection of colon cancer in the Second People’s Hospital of Yunnan province from 2017 to 2018 was detected by immunohistochemistry.The interaction between C/EBPδand Siah2 in colorectal cancer tissues was detected by immunoprecipitation.Viral vectors with C/EBPδoverexpression and interference were constructed to infect colorectal cancer caco-2 cells,respectively.The effects of C/EBPδexpression and interference on cell proliferation,apoptosis and cycle were observed through CCK8,flow cytometry detection and plate cloning formation experiments.Chi-square test was used to analyze the correlation factors of positive expression rate.ResultsⅠThe expression of C/EBPδincreased successively from the normal intestinal tissues to paracellular tissues and then to colorectal cancer tissues,and the differences were statistically significant(all P<0.05).The expression of C/EBPδprotein was more significant in colorectal cancer patients with liver metastasis;ⅡThe C/EBPδincreased colorectal cancer tissues were accompanied by the increase of Saih2,and there was an interaction between the two;ⅢC/EBPδoverexpression significantly increased the proliferation of caco-2 cells,while the shRNA interference of C/EBP significantly reduced the proliferation of colorectal cancer cells;ⅣCompared with the control group,the shRNA interference of C/EBPδsignificantly inhibited the in vitro migration and invasion of colorectal cancer cells caco-2.Conclusion C/EBPδis abnormal in colorectal cancer tissues,and the overexpressed C/EBPδpromotes the invasion and rotation of colorectal cancer cells by interacting with Saih2,which may be related to the poor prognosis of colorectal cancer.
