摘  要:为评估浙江省三门湾海域鱼类种质资源现状,采用简化基因组测序技术,对三门湾海域3种优势鱼种(白姑鱼Pennahia argentata、中华栉孔虾虎鱼Ctenotrypauchen chinensis和龙头鱼Harpadon nehereus)进行群体遗传多样性分析。结果表明,白姑鱼、中华栉孔虾虎鱼、龙头鱼获得单核苷酸多态性(SNP)位点分别为125225个、51283个、55128个,变异位点比例分别为1.74%、0.79%、0.54%,多态信息含量(PIC)分别为0.208、0.177、0.221,核苷酸多样性(P_(i))分别为0.259、0.219、0.281。其中白姑鱼观测杂合度小于期望杂合度,显示出群体内纯合子较多,杂合子缺失;而中华栉孔虾虎鱼、龙头鱼观测杂合度大于期望杂合度,显示出杂合过度。3种鱼类群体以龙头鱼观测杂合度最高。综合以上SNP位点多样性、多态性信息含量、群体遗传多样性比较结果,三门湾海域3种鱼类群体变异位点比例均较低,相同群体的个体间遗传分化较小,遗传结构较稳定,总体上遗传多样性处于较低水平,应进一步加强资源保护和恢复。In order to evaluate the status of germplasm resources in the Sanmen Bay,Zhejiang Province,this study conducted population genetic diversity analysis of three dominant fishes in the Sanmen Bay(Pennahia argentata,Ctenotrypauchen chinensis,and Harpadon nehereus)from 2019 to 2020 based on simplified genome sequencing technology.The results of genetic analysis showed that the number of single nucleotide polymorphism(SNP)sites obtained by P.argentata,C.chinensis,and H.nehereus were 125225,51283 and 55128,and the proportions of variable sites were 1.74%,0.79%,and 0.54%,respectively.The polymorphism information content(PIC)was 0.208,0.177 and 0.221,and the nucleotide diversity(P_(i))was 0.259,0.219 and 0.281.The observed heterozygosity of P.argentata was less than the expected heterozygosity,showing that there were more homozygotes and fewer heterozygotes in the population;while the observed heterozygosity of C.chinensis and H.nehereus was greater than the expected heterozygosity,showing excessive heterozygosity.Among three fish species,H.nehereus had the highest heterozygosity.Based on the above single nucleotide polymorphism sites diversity,polymorphism information content,and population genetic diversity comparison results,the three dominant fish populations sampled in the Sanmen Bay sea area had a low proportion of mutation sites,the genetic differentiation among individuals of the same population was small,the genetic structure was relatively stable,and the genetic diversity level was low.In the future,protection and restoration should be strengthened.
