平消胶囊含药血清联合紫杉醇对乳腺癌细胞的作用及机制研究

摘　　要：目的:研究平消胶囊含药血清联合紫杉醇对乳腺癌细胞的作用及机制。方法:用平消胶囊灌喂大鼠以制备平消胶囊含药血清,分别用无药血清、平消胶囊含药血清、紫杉醇、平消胶囊含药血清联合紫杉醇处理乳腺癌MCF-7细胞株,分别依次为对照组、含药血清组、紫杉醇组、联合组。用MTT实验检测药物对乳腺癌细胞的抑制效果;用流式细胞仪检测细胞凋亡情况及周期变化;采用蛋白印迹法检测Bax、Bcl-2、Caspase-3、AMPK和P21蛋白的表达量。结果:与对照组相比,含药血清组、紫杉醇组和联合组中细胞增殖力均被显著抑制,但联合组更明显(P<0.05)。流式细胞仪检测结果显示,与对照组相比,含药血清组、紫杉醇组和联合组肿瘤细胞周期G0/G1发生阻滞,细胞凋亡率增大,但联合组效果更显著(P<0.05)。与对照组相比,含药血清组、紫杉醇组和联合组中Bcl-2蛋白均显著下调(P<0.05),Bax和Caspase-3、AMPK和P21蛋白上调,联合组的效果更显著(P<0.05)。结论:平消胶囊含药血清联合紫杉醇可能通过激活AMPK信号通路,诱导凋亡,从而抑制乳腺癌细胞增殖。Objective: To study the effect and mechanism of pingxiao-capsule-containing-serum combined with paclitaxel on breast cancer cells. Methods: Rats were fed with Pingxiao capsules to prepare drug-containing serum of Pingxiao capsules. MCF-7 cell lines were treated with drug-free serum(control group), drug-containing serum of Pingxiao capsules(drug-containing serum group), paclitaxel group, and drug-containing serum of Pingxiao capsules combined with paclitaxel(combination group), respectively. MTT assay was used to detect the inhibitory effect of drugs on breast cancer cells, and flow cytometry was used to detect cell apoptosis and cell cycle.Western blotting was used to detect the expression of Bax, Bcl-2, Caspase-3, AMPK and P21 proteins. Results:Compared with the control group, the G0/G1 cell cycle was blocked in the drug-containing serum group, paclitaxel group and combination group, the cell proliferation and Bcl-2 protein expression level were decreased, while the apoptosis rate and the protein expression levels of Bax, Caspase-3, AMPK and P21 were increased(P<0.05).These changes were the most significant in the combination group(P<0.05). Conclusion: Pingxiao-capsule-containing-serum combined with paclitaxel can inhibit the proliferation of breast cancer cells by activating AMPK signaling pathway and promoting cell apoptosis.
