摘 要:目的研究黄绿青霉素(CIT)诱导神经细胞PC-12的损伤作用。方法CIT处理组分为三组,终浓度分别为2.5μmol/L、5μmol/L、10μmol/L,设DMSO为空白对照组。药物处理细胞24h后,观察细胞形态的变化,CCK-8法测细胞活性,流式细胞仪测ROS荧光值;取细胞培养液测LDH释放率、NO含量及MDA;细胞裂解液检测GSH含量及SOD活力。结果PC-12与CIT共培养后,细胞成团,出现空斑。与对照组比较,CIT组的细胞活力显著降低(P<0.01),LDH释放率显著升高(P<0.01);NO含量明显下降(P<0.05),呈剂量效应关系;氧化应激相关指标MDA含量显著升高(P<0.01);ROS含量在低、中、高剂量组中均显著升高(P<0.01,P<0.01,P<0.05);GSH含量呈下降趋势,10μmol/L组效果显著(P<0.05);SOD活力显著升高(P<0.01)。结论黄绿青霉素对神经细胞PC-12有明显的损伤作用,可能与氧化应激机制有关。Objective To investigate the injury of PC-12 induced by Citreoviridin(CIT).Methods PC-12 cells were treated with different concentrations of Citreoviridin,which were 2.5μmol/L,5μmol/L and 10μmol/L respectively,the control group was treated with DMSO.After treated with CIT for 24 hours,cell morphology was observed,cell activity was measured by CCK-8 method,ROS fluorescence was measured by flow cytometry.LDH release rate,NO content and MDA were measured with cell culture medium,GSH content and SOD activity were measured with cell lysate.Results After the co culture of PC-12 and CIT,the cells shrunk into clusters and appeared empty spots.Compared with the control group,the cell viability of CIT group decreased significantly(P<0.01),and the LDH release rate increased significantly(P<0.01).NO content decreased significantly(P<0.05),showing a dose-response relationship.MDA content increased significantly(P<0.01);ROS content in all groups had significant effect(P<0.01,P<0.01,P<0.05);GSH content decreased,10μmol/L group had a significant effect(P<0.05);SOD activity increased significantly(P<0.01).Conclusion Citreoviridin can damage PC-12 cells effectively,which may be induced by oxidative stress mechanism.
