摘 要:维生素D_(3)(Vitamin D_(3),VD_(3))和染料木素(Genistein,Gen)二者均具有抗骨质疏松作用,然而,二者协同作用机制及维生素D_(3)的抗骨质疏松作用使用剂量有待阐明。本研究报告了维生素D_(3)和染料木素对人成骨肉瘤细胞MG-63、小鼠巨噬细胞RAW264.7诱导的破骨细胞增殖与分化的影响机制。维生素D_(3)联合染料木素可显著促进成骨细胞的增殖分化,促进骨保护素(OPG)的表达,抑制核因子κB(NF-κB)受体激活剂(RANKL)的表达,提高OPG和RANKL mRNA表达比值。此外,维生素D_(3)在6.25~50.00μmol/L剂量范围,可促进破骨细胞的增殖和成熟,而联合染料木素后表现为显著抑制作用。维生素D_(3)可促进破骨细胞中组织蛋白酶K的表达,联合染料木素处理细胞,显著抑制酒石酸抗性磷酸酶(TRAP)和组织蛋白酶K的表达。试验结果表明,维生素D_(3)联合染料木素,可显著促进成骨细胞的增殖和分化。维生素D_(3)表现为浓度依赖的增强破骨细胞增殖和成熟作用,而联合染料木素可显著抑制破骨细胞增殖和成熟,间接起到抗骨质疏松作用。Vitamin D_(3) is a fat-soluble vitamin synthesized endogenously in the skin,and genistein is an isoflavone mainly found in legumes.Both have anti-osteoporosis effects,and the two compounds combination also promotes bone formation.However,the synergistic mechanism of the two and the dosage of anti-osteoporosis effect of vitamin D_(3) need to be clarified.In this study,the effect of vitamin D_(3) and genistein on the proliferation and differentiation of osteoclasts induced by human osteosarcoma cell MG-63 and mouse macrophage RAW264.7 was reported.The osteoblast(MG-63)proliferation was increased significantly by VD_(3) and genestein combination,and osteoblast indicators of alkaline phosphatase(ALP)activity,calcium nodules,the expression of osteoprotegerin(OPG),ratio of OPG and receptor activator of nuclear factor kappa B(NF-κB)ligand(RANKL)in mRNA level were also promoted significantly in combination group,compared with both single groups.In addition,the proliferation and maturation of osteoclasts(RAW264.7 RANKL induced)were promoted by VD_(3) with 6.25-50.00μmol/L dosage,but inhibited significantly by combination of VD_(3) and genistein.Furthermore,the expression of cathepsinK mRNA in osteoclasts was promoted by VD_(3),but the expression of tartrate resistant phosphatase(TRAP)and CathepsinK mRNA were inhibited in the combination group.These results showed that the proliferation and differentiation of osteoblasts could be significantly promoted by vitamin D_(3) combined with genistein,indicating bone density increase directly.Vitamin D_(3),showed a dose dependent promoting effect on osteoclast proliferation and maturation,could inhibit osteoclast proliferation and maturation significantly,playing an anti-osteoporosis role indirect after combined with genistein.
