摘 要:目的建立同时测定豆板还阳药材中3种成分含量的高效液相色谱法。方法色谱柱为岛津InertSustain C18柱(150 mm×4.6 mm,5μm),流动相为乙腈-0.1%磷酸水溶液(梯度洗脱),流速为1.0 mL/min,检测波长为332 nm,柱温为30℃,进样量为10μL。采用SPSS 21.0统计学软件对8批不同产地豆板还阳药材进行聚类分析。结果阿魏酸、槲皮素、石吊兰素在各自质量浓度范围内与峰面积线性关系良好(R2≥0.9997);精密度、稳定性、重复性试验的RSD≤2.30%;平均加样回收率在101.37%~102.54%范围内,RSD为2.94%~3.27%(n=6);8批药材中阿魏酸、槲皮素、石吊兰素总含量介于0.147%~0.553%。经聚类分析,8批药材可分为三类。结论该方法操作简便,结果准确,重复性和稳定性均好,可用豆板还阳药材中3种成分的含量测定,且3种成分在不同产地的药材中含量差异较大。Objective To establish a high-performance liquid chromatography(HPLC)method for the simultaneous determination of three components in Lysionoti Herba.Methods The chromatographic column was InertSustain C18 column(150 mm×4.6 mm,5μm),the detection wavelength was 332 nm,the mobile phase was acetonitrile-0.1%phosphoric acid aqueous solution(gradient elution),the flow rate was 1.0 mL/min,the column temperature was 30℃,a nd the injection volume was 10μL.SPSS 21.0 statistical software was used to cluster analysis of eight batches of Lysionoti Herba from different origins.Results The ferulic acid,quercetin and lysionotin showeda good relationship with the peak area in their respective mass concentration range(R2≥0.9997).The RSDs of precision,stability and repeatability tests were not more than 2.30%.The average recoveries of ferulic acid,quercetin and lysionotin were in the range of 101.37%-102.54%with RSD of 2.94%-3.27%(n=6).The total content of ferulic acid,quercetin and lysionotin in eight batches of medicinal materials was in the range of 0.147%-0.553%.Cluster analysis showed that eight batches of medicinal materials could be divided into three categories.Conclusion The method is simple,accurate,reproducible and stable,which can be used for the content determination of three components in Lysionoti Herba,and the content of the three components varies greatly in Lysionoti Herba from different producing areas.
