摘　　要：目的观察结肠癌患者血清长链非编码RNA(lncRNA)锌指蛋白反义链1(ZFAS1)水平变化,并探讨其临床意义。方法选择102例结肠癌患者(结肠癌组)和54例健康志愿者(对照组),采用实时荧光定量PCR技术检测血清lncRNAZFAS1,分析lncRNAZFAS1水平与结肠癌患者临床病理特征以及预后的关系。结果结肠癌组血清lncRNAZFAS1水平高于对照组(P<0.01)。血清lncRNAZFAS1水平与结肠癌分化程度、T分期、N分期有关(P均<0.01)。Kaplan-Meier生存曲线分析结果显示,lncRNAZFAS1高水平者3年无进展生存(PFS)率、总生存(OS)率低于lncRNAZFAS1低水平者(P均<0.05)。多因素Cox比例风险回归分析显示,N1~N2期、高水平lncRNAZFAS1是结肠癌患者不良预后的危险因素(P均<0.01)。结论结肠癌患者血清lncRNAZFAS1水平升高,lncRNAZFAS1水平与结肠癌分化程度、T分期、N分期和预后有关,可作为结肠癌预后评估的潜在标志物。Objective To observe the expression changes of long non-coding RNA(lncRNA)Zinc finger antisense 1(ZFAS1)in serum of colon cancer patients and its clinical significance.Methods Totally 102 patients with colon cancer(colon cancer group)and 54 volunteers(control group)were selected.Serum lncRNA ZFAS1 expression was detected by real-time fluorescence quantitative polymerase chain reaction(qRT-PCR).We analyzed the relationship between lncRNA ZFAS1 expression and clinicopathological features and prognosis of colon cancer patients.Results The relative expression of lncRNA ZFAS1 in serum of the colon cancer group was higher than that of the control group(P<0.01).Expression of lncRNA ZFAS1 was related to the degree of differentiation,T stage and N stage of colon cancer(all P<0.01).Kaplan-Meier survival curve analysis showed that the 3-year progression-free survival(PFS)and overall survival(OS)of patients with high lncRNA ZFAS1 expression were lower than those with low lncRNA ZFAS1 expression(both P<0.05).Multivariate Cox proportional risk regression analysis showed that N1-N2 and high expression of lncRNA ZFAS1 were risk factors for poor prognosis of patients with colon cancer(both P<0.01).Conclusions The expression of lncRNA ZFAS1 in serum increases in patients with colon cancer.The expression of lncRNA ZFAS1 is related to the degree of differentiation,T stage,N stage and prognosis of colon cancer,and can be used as a potential marker for prognosis evaluation of colon cancer.
