摘 要:目的分析沉默信息调节因子6(SIRT6)与上皮细胞钙黏蛋白(E-cadherin)在食管鳞癌组织及同一患者的癌旁正常组织中的表达及其临床意义。方法选取2018年6月至2019年12月郑州大学附属洛阳中心医院胸外科收治的61例手术治疗食管鳞癌患者的癌组织及距肿瘤切缘3 cm以上的癌旁组织(镜下观察未见癌细胞),男42例,女19例,年龄(63.62±9.59)岁,年龄范围为34~82岁。通过免疫细胞化学与蛋白免疫印迹实验(Western blot)方法检测SIRT-6、E-cadherin在食管鳞癌组织中的蛋白表达及免疫组化,并分析SIRT6和E-cadherin与临床病理资料的相关性。结果在食管鳞癌中SIRT6蛋白相对表达量(0.383±0.092)显著低于癌旁组织(1.037±0.116),差异有统计学意义(t=9.877,P<0.01)。食管鳞癌中E-cadherin蛋白相对表达量(0.423±0.089)低于癌旁组织(0.967±0.116),差异有统计学意义(t=6.976,P<0.01)。SIRT6在食管鳞癌组织中阳性表达与浸润深度相关,差异有统计学意义(P<0.05);与肿瘤大小、分化程度、淋巴结转移无关,差异无统计学意义(P>0.05)。E-cadherin在食管鳞癌组织中阳性表达与分化程度、浸润深度相关,差异有统计学意义(P<0.05);与肿瘤大小、淋巴结转移无关,差异无统计学意义(P>0.05)。进行Spearman等级相关分析显示,两者之间呈正相关关系,差异有统计学意义(r=0.538,P<0.01)。结论SIRT-6和E-cadherin的表达影响食管鳞癌的侵袭性,可为临床治疗食管鳞癌提供理论依据及靶点。Objective To analyze the correlation between silent information regulator 6(SIRT6)and epithelium-Ca2+dependent cell adhesion molecule(E-cadherin)in the esophageal squamous cell carcinoma and adjacent normal tissues of the same patient.Methods A total of 61 patients with esophageal squamous cell carcinoma(ESCC)who received surgical treatment in the department of Thoracic Surgery of Luoyang Central Hospital Affiliated to Zhengzhou University from June 2018 to December 2019 were selected from the sample,including 42 males and 19 females,aged(63.62±9.59)years old,ranging from 34 to 82 years old,whose cancer tissues and paracancerous tissues more than 3 cm from the margin of tumor resection(no cancer cells observed under microscope).The protein expression and immunohistochemistry of SIRT-6 and E-cadherin in esophageal squamous cell carcinoma were detected by immunocytochemistry(IHC)and Western blot assay.The correlation of SIRT6 and E-cadherin with clinicopathological data was analyzed.Results The relative expression level of SIRT6 protein in esophageal squamous cell carcinoma(0.383±0.092)was significantly lower than that in adjacent tissues(1.037±0.116),and the difference was statistically significant(t=9.877,P<0.01).The relative expression level of E-cadherin protein(0.423±0.089)was significantly lower than that in adjacent tissues(0.967±0.116),and the difference was statistically significant(t=6.976,P<0.01).The positive expression of SIRT6 in esophageal squamous cell carcinoma was correlated with the depth of invasion,and the difference was statistically significant(P<0.05).There was no significant difference in tumor size,degree of differentiation and lymph node metastasis(P>0.05).The positive expression of E-cadherin in esophageal squamous cell carcinoma was correlated with differentiation degree and invasion depth,and the difference was statistically significant(P<0.05).There was no significant difference in tumor size and lymph node metastasis(P>0.05).Spearman rank correlation analysis showed that there wa
