摘 要:目的:探讨磷脂酰肌醇3激酶/心肌素(PI3K/MYOCD)信号通路在调控血管平滑肌瘤(AM)细胞中的作用。方法:AM细胞分为PI3K/MYOCD siRNA组(转染PI3K/MYOCD siRNA)和对照组(未行转染)。qRT-PCR检测PI3K和MYOCD mRNA的表达,CCK8试剂盒检测细胞的增殖能力,Transwell小室检测细胞的侵袭能力,Western blot检测PI3K、MYOCD、AKT、Bcl-2蛋白的表达。结果:PI3K/MYOCD siRNA组细胞的增殖和侵袭能力低于对照组(P<0.05);与对照组相比,PI3K/MYOCD siRNA组细胞的PI3K、MYOCD表达水平降低,AKT蛋白表达水平降低,Bcl-2蛋白表达水平升高(P<0.05);PI3K/MYOCD siRNA组细胞的PI3K蛋白与MYOCD、AKT蛋白呈正相关,与Bcl-2蛋白呈负相关;MYOCD蛋白与AKT蛋白呈正相关,与Bcl-2蛋白呈负相关(P<0.001)。结论:抑制PI3K/MYOCD信号通路可能通过调控AKT、Bcl-2蛋白降低AM细胞的增殖和侵袭能力。Aim:To investigate the effects and mechanism of inhibiting the phosphatidylinositol 3-kinase/myocardin element(PI3K/MYOCD)signaling pathway on the regulation of angioleiomyoma(AM)cells.Methods:AM cells were divided into PI3K/MYOCD siRNA group(transfected with PI3K/MYOCD siRNA)and control group(not transfected).qRT-PCR was used to detect the expressions of PI3K and MYOCD mRNA,CCK8 kit was used to detect the proliferation of the cells,Transwell chamber was used to detect the invasion of the cells,and Western blot was used to detect the expressions of PI3K,MYOCD,AKT and Bcl-2 proteins.Results:The proliferation and invasion of the cells in PI3K/MYOCD siRNA group were lower than those in control group(P<0.05).Compared with the control group,the expression levels of PI3K,MYOCD,and AKT proteins decreased,and that of Bcl-2 protein increased in PI3K/MYOCD siRNA group(P<0.05);in PI3K/MYOCD siRNA group,the expression level of PI3K protein was positively correlated with those of MYOCD and AKT proteins,and negatively correlated with that of Bcl-2 protein;the expression level of MYOCD protein was positively correlated with that of AKT protein and negatively correlated with that of Bcl-2 protein(P<0.001).Conclusion:Inhibition of PI3K/MYOCD signaling pathway may inhibit the proliferation and invasion of AM cells by regulating AKT,Bcl-2 and other proteins.
