摘 要:目的:利用生物信息学分析来识别肝细胞癌(HCC)组织与正常肝组织的差异表达基因,并寻找潜在的诊断分子标志物和治疗靶点。方法:从GEO数据库搜索并下载数据集,通过GEO2R在线分析工具计算差异表达基因,使用韦恩图在线工具识别数据集共同的差异表达基因。对这些共同的差异表达基因用DAVID在线工具进行GO和KEGG通路富集分析,利用Cytoscape软件构建蛋白相互作用网络,并选出关键基因(度值排名前十的基因)。使用GEPIA数据库进行生存分析,评估关键基因与HCC预后的关系。结果:从选出的4个数据集(GSE62232、GSE57957、GSE121248和GSE39791)中鉴定出150个共同的差异表达基因,其中26个表达上调,124个表达下调。KEGG通路分析表明“代谢通路”“碳代谢”“视黄醇代谢”“矿物质吸收”和“补体系统”是重要的通路。细胞色素P4503A4酶(CYP3A4)和酪氨酸转氨酶(TAT)基因与HCC的生存有关。结论:CYP3A4和TAT基因可能成为HCC的诊断分子标志物和治疗靶点。Aim:To identify differentially expressed genes(DEG)between hepatocellular carcinoma(HCC)tissue and normal tissue by bioinformatics analysis,so as to search for potential diagnostic molecular markers and therapeutic targets.Methods:Datasets from the Gene Expression Omnibus(GEO)database were searched and downloaded.The DEG were analyzed by GEO2R online analysis tool.The intersection DEG were identified by the Venn diagram webtool.GO and KEGG pathway analysis were used to explore the function of genes.The protein-protein interaction network for intersection DEG was constructed by the Cytoscape software,and ten genes were selected.The GEPIA database was used to make survival analysis and evaluate the prognostic value of hub genes in HCC patients.Results:A total 150 DEG were selected from 4 qualified datasets(GSE62232,GSE57957,GSE121248 and GSE39791),including 26 upregulated genes and 124 downregulated genes.The KEGG pathway analysis demonstrated that"metabolic pathways""carbon metabolism""retinol metabolism""mineral absorption"and"complement and coagulation cascades"were the significant pathways.CYP3A4 and TAT were associated with overall survival of HCC patients.Conclusion:CYP3A4 and TAT genes may become diagnostic molecular markers and therapeutic targets for HCC.
