摘  要:本研究以牛源链球菌48株为宿主,经双层琼脂平板反复纯化从污水中分离到毒性噬菌体SP48。透射电镜观察发现,SP48属于长尾噬菌体科;酶切分析表明,SP48的基因组大于60 kb,可被限制性核酸内切酶Eco R I、Xho l I和Bln I切开。对乙醚、氯仿不敏感,对40~45℃或pH 5~10的环境有很好的耐受性;最佳感染复数为10,潜伏期约30 min,爆发期约80 min,平均爆发量为45 PFU/cell。In this study,we research used Streptococcus 48 strain isolated from cow uterus of endometritis,to isolate the virulent bacteriophage SP48 from sewsage by double agar purification method.Using transmission electron microscopic observation,we found that SP48 belonged to Siphoviridae,and the enzyme digestion analysis indicated that its genome was bigger than 60 kb and could be digested by restriction enzymes Eco R I,Xho l I and Bln I.The results of biological characteristics studies showed that ether or chloroform hardly destroyed the infectivity of SP48.SP48 had excellent tolerance to 40-45℃,pH 5-10.The optimal multiplicity of infection was 10,the latent period was 30 min,the burst period was 80 min,and the average burst size was about 45 PFU/cell.
