钝齿蟳线粒体基因组全序列测定及系统发育分析

摘　　要：线粒体基因组全序列为更好地理解分子进化和系统发育关系提供了重要信息。本研究首次测定了钝齿蟳的线粒体基因组全序列,其全长为15910 bp,包括13个蛋白编码基因,22个tRNA基因,2个rRNA基因和1个控制区。整个线粒体基因组的核苷酸组成为33.8%A、36.4%T、11.1%G及18.7%C,具有明显的AT偏向性(70.2%)。所有蛋白编码基因以ATN作为起始密码子,绝大多数蛋白编码基因以TAG或TAA作为终止密码子,少数以不完全密码子(T-)作为终止。除tRNA-Ser1和tRNA-Thr分别缺失二氢尿嘧啶臂(DHU臂)和TΨC环外,其余所有的tRNA基因均能形成典型的三叶草结构。系统发育分析结果显示,蟳属所有物种都聚为一个类群,并与短桨蟹属有着最近的亲缘关系。这些结果将有助于更好地了解钝齿蟳线粒体基因组序列特征,并为研究梭子蟹科的系统发育关系奠定基础。Complete mitochondrial genome(mitogenome)can provide important information for better understanding of molecular evolution and phylogenetic analysis.In this study,the complete mitogenome sequence of Charybdis hellerii(Brachyura:Portunidae)was first determined.The total length is 15910 bp and includes 13 protein-coding genes(PCGs),22 transfer RNAs(tRNAs),two ribosomal RNAs(rRNAs),as well as a putative control region(CR).The nucleotide composition of the whole mitogenome is 33.8%A,36.4%T,11.1%G,and 18.7 C;respectively,which reveals a strong AT bias(70.2%).Except tRNA-Ser1 and tRNA-Thr,other tRNAs are able to form the typical clover structure.Phylogenetic analysis show that all Charybdis species are placed into one group,and form a sister clade with Thalamita.These results will help to better understand the molecular features of the C.hellerii mitogenome and lay a foundation for phylogenetic relationships within Portunidae.
