静脉留置针致静脉血栓形成对白兔耳缘静脉中血管内皮损伤标志物表达水平的影响

摘　　要：目的探讨静脉留置针致静脉血栓形成对白兔耳缘静脉中血管内皮生长因子(VEGF)、血管假性血友病因子(vWF)、组织型纤溶酶原激活物(t-PA)表达水平的影响。方法将70只新西兰大白兔随机分为空白组(n=10)和置管组(n=60),再将置管组分为生理盐水组、甘露醇组各30只。于置管组白兔耳缘静脉留置静脉留置针后滴注相应药物,空白组不做任何处理。分别于置管后1 d、3 d、5 d取各组白兔耳缘静脉标本,镜下观察血栓形成情况,并检测VEGF、vWF、t-PA表达情况。结果(1)置管后1 d,在生理盐水组、甘露醇组白兔血管标本中均可观察到血栓形成。(2)各时间点,3组VEFG表达水平差异均无统计学意义(P>0.05)。(3)置管后1、3 d,各组vWF表达水平差异均无统计学意义(P>0.05);置管后5 d,生理盐水组、甘露醇组vWF表达水平均低于空白组及同组置管后1 d的水平(P<0.05)。(4)置管后1、5 d,各组t-PA表达水平差异均无统计学意义(P>0.05)。但置管后3 d,生理盐水组t-PA表达水平高于空白组(P<0.05)。结论留置静脉留置针致静脉血栓形成后,白兔耳缘静脉中vWF表达水平有一定的变化,但VEGF、t-PA表达水平变化不明显。Objective To investigate the effect of venous thrombosis induced by indwelling venous catheter on the expression levels of vascular endothelial growth factor(VEGF),von Willebrand factor(vWF)and tissue-type plasminogen activator(t-PA)in ear vein of white rats.Methods Seventy New Zealand white rabbits were randomly divided into blank group(n=10)and catheterization group(n=60),and the catheterization group was subdivided into normal saline group(n=30)and mannitol group(n=30).Rabbits in the catheterization group received infusion of corresponding drugs after indwelling venous catheter via ear vein,and the blank group did not receive any treatment.The ear vein samples of white rabbits were collected from each group on days 1,3,and 5 after catheterization,respectively,to observe the condition of thrombosis under microscope and detect the expression of VEGF,vWF and t-PA.Results(1)One day after catheterization,thrombosis was observed in the venous samples of the white rabbits in the normal saline and mannitol groups.(2)There was no statistically significant difference in VEGF expression level among the three groups at any time point(P>0.05).(3)The expression levels of vWF on days 1 and 3 after catheterization showed no statistically significant difference in each group(P>0.05).The expression level of vWF on day 5 after catheterization in the normal saline and mannitol groups was lower than that in the blank group,and also lower than the level on day 1 after catheterization(P<0.05).(4)The expression levels of t-PA on days 1 and 5 after catheterization showed no statistically significant difference in each group(P>0.05).However,three days after catheterization,the normal saline group yielded a higher expression level of t-PA than the blank group(P<0.05).Conclusion After venous thrombosis induced by indwelling venous catheter,the expression level of white rabbit ear vein vWF exhibits changes to some extent,but the expression level of VEGF or t-PA does not change significantly.
