摘  要:SRPX2是一种具有细胞外基质蛋白属性的硫酸软骨素蛋白多糖,该基因最早发现于白血病细胞中,后来进一步的研究发现其同时也具有参与癫痫发作、血管生成、细胞黏附等多种生物学行为。该基因正常表达于脑神经元、心、肺、气管等多种正常的组织中,该基因发生突变会导致癫痫发作、言语缺失等,影响正常神经系统的发育。后续研究发现该基因同时具有促血管生成、细胞黏附的作用。多项相关研究报道SRPX2参与了早期的血管生成,并主要通过与uPAR的相互作用参与血管生成早期阶段的血管内皮结构重塑。研究发现新生血管内皮细胞中存在SRPX2和uPAR共表达,通过siRNA沉默SRPX2基因可以特异性抑制小鼠t.End.1V内皮细胞迁移及早期出芽的形成;SRPX2高表达可激活整合素αvβ3/FAK信号通路参与调节血管内皮细胞迁移、黏附等生物学行为,并具有促管腔形成作用。深入研究其在促进血管生成过程中的相关调节因素,为后期进一步研究通过体外基因转染优化内皮细胞结构、生物学功能,促进缺血组织血管新生,对防治下肢缺血性疾病具有重要参考意义。Srpx2 is a kind of chondroitin sulfate proteoglycan with the properties of extracellular matrix protein.The gene was first found in leukemia cells.Later,further studies found that srpx2 is also involved in epilepsy,angiogenesis,cell adhesion and other biological behaviors.The gene is expressed in brain neurons,heart,lung,trachea and other normal tissues.The mutation of the gene is related to epilepsy,speech loss and so on,which affects the development of normal nervous system.It was found that the gene could promote angiogenesis and cell adhesion.Anumber of related studies have reported that srpx2 is involved in early angiogenesis,and mainly through the interaction with uPAR involved in vascular endothelial structure remodeling in the early stage of angiogenesis.It was found that srpx2 and uPAR were co-expressed in neovascular endothelial cells.Silencing srpx2 gene by siRNA can specifically inhibit the migration and early budding of mouse t.End.1V endothelial cells;high expression of srpx2 can activate integrinαVβ3/FAK signaling pathway,participate in regulating the migration and adhesion of vascular endothelial cells,and promote the formation of lumen.Further study of its regulatory factors in the process of promoting angiogenesis has important reference significance for further study of optimizing the structure and biological function of endothelial cells through in vitro gene transfection,promoting angiogenesis of ischemic tissue,and preventing and treating lower limb ischemic diseases.
