摘 要:目的:研究经低氧预适应处理的血管内皮细胞分泌的物质对神经细胞低氧耐受的影响。方法:分别将人微血管内皮细胞(HMEC-1)经过低氧预适应(HPC)处理及正常培养,分离其培养基并通过滤膜过滤。人神经母细胞瘤细胞系(SH-SY5Y)细胞分为低氧预适应组(HPC组)、培养基对照组(Medium-C组)和空白对照组(SY5Y-C组),三组细胞分别加入经HPC的HMEC-1细胞的培养基、正常培养HMEC-1细胞的培养基和正常培养SH-SY5Y细胞的培养基。随后三个组细胞均给予低氧处理。使用MTS法检测神经细胞活力,Western Blot检测caspase-3蛋白表达水平。结果:SY5Y-C组、Medium-C组和HPC组的细胞活力分别是(0.9967±0.0033)、(1.0751±0.0447)和(1.4737±0.0325),HPC组细胞活力显著增加(P<0.05);三个组caspase-3蛋白表达的相对丰度分别是(0.9950±0.0050)、(0.9893±0.0787)、(0.8180±0.0361),HPC组细胞的caspase-3蛋白表达显著受到抑制(P<0.05)。结论:内皮细胞在经过低氧预适应处理后分泌的物质可以增加神经细胞的低氧耐受。Objective:To explore the effect of substances secreted by vascular endothelial cells treated with hypoxia preconditioning on the hypoxia tolerance of nerve cells. Methods:Human microvascular endothelial cells(HMEC-1) were treated with hypoxic preconditioning(HPC) and cultured normally. Then the cultured medium was separated and filtered through a filter membrane. Human neuroblastoma cell line(SH-SY5 Y) cells were divided into hypoxic preconditioning group(HPC group), medium control group(HMEC-1 group) and blank control group(SY5 Y-C group). The cells in the three groups were cultured with medium from HPC-treated HMEC-1 cell, normal cultured HMEC-1 cells, and normal cultured SH-SY5 Y medium. Then the cells in the three groups were given hypoxia treatment. The MTS method was used to detect nerve cell viability, and Western Blot was used to detect the expression level of caspase-3 protein. Results:The cell viability of SY5 Y-C group, HMEC-1 group and HPC group were(0.9967±0.0033),(1.0751±0.0447) and(1.4737±0.0325), respectively. The cell viability of HPC group increased significantly(P<0.05);The relative abundances of caspase-3 protein expression in each group were 0.9950±0.0050, 0.9893±0.0787 and 0.8180±0.0361. The caspase-3 protein expression of cells in the HPC group was significantly inhibited(P<0.05). Conclusion:The substances secreted by endothelial cells after hypoxic preconditioning can increase the hypoxic tolerance of nerve cells.
