摘 要:目的研究布比卡因对胆囊癌GBC-SD细胞增殖、侵袭和微管形成的影响。方法采用CCK-8法检测不同浓度布比卡因对胆囊癌GBC-SD细胞活性的影响;根据结果将GBC-SD细胞分为对照组、1.25、2.5和5 mmol/L组,对照组不用布比卡因处理,1.25、2.5和5 mmol/L组分别应用1.25、2.5和5 mmol/L布比卡因处理。克隆形成实验、Transwell实验和微管形成实验分别检测胆囊癌GBC-SD细胞增殖、侵袭及微管形成结节数;采用Western blot检测p21、Survivin、Bax、Bcl-2、E-cadherin、N-cadherin、Vimentin和血管内皮生长因子(VEGF)蛋白表达量。结果与对照组和1.25 mmol/L组比较,2.5 mmol/L组和5 mmol/L组细胞克隆形成比率、细胞侵袭力、微管形成结节数及Survivin、N-cadherin、Vimentin、VEGF蛋白表达量显著下调,p21、E-cadherin蛋白表达量和Bax/Bcl-2比率显著上调(P<0.05,P<0.01)。与2.5mmol/L组比较,5 mmol/L组细胞克隆形成比率、细胞侵袭力、微管形成结节数及Survivin、N-cadherin、Vimentin、VEGF蛋白表达量显著下调,p21、E-cadherin蛋白表达量和Bax/Bcl-2比率显著上调(P<0.05)。结论布比卡因能够抑制胆囊癌GBC-SD细胞的增殖、侵袭和微管形成。Objective To investigate the effects of Bupivacaine on proliferation,invasion and microtubule formation of GBC-SD cells in gallbladder cancer.Methods CCK-8 method was used to detect the effects of different concentrations of Bupivacaine on the activity of GBC-SD cells in gallbladder cancer.According to the results,GBC-SD cells were divided into control group,1.25 mmol/L,2.5 mmol/L and 5 mmol/L Bupivacaine groups.Control group was not treated with Bupivacaine,while the 1.25,2.5 and 5 mmol/L Bupivacaine groups were treated with 1.25,2.5 and 5 mmol/L Bupivacaine respectively.Conditions of proliferation,invasion and the number of microtubule nodules of GBC-SD cells were detected by clonal formation assay,Transwell assay and microtubule-forming assay respectively.Protein expressions of p21,Survivin,Bax,Bcl-2,E-cadherin,N-cadherin,Vimentin and vascular endothelial growth factor(VEGF)were detected by Western Blot.Results Compared with those in control group and 1.25 mmol/L Bupivacaine group,clonal formation rate of cells,cell invasion,number of microtubule nodules and protein expressions of Survivin,N-cadherin,Vimentin and VEGF in GBC-SD cells were significantly down-regulated,while protein expressions of p21 and E-cadherin,and Bax/Bcl-2 ratio were significantly up-regulated in 2.5 mmol/L and 5 mmol/L Bupivacaine groups(P<0.05,P<0.01).Compared with those in 2.5 mmol/L Bupivacaine group,clonal formation rate of cells,cell invasion,number of microtubule nodules and protein expressions of Survivin,N-cadherin,Vimentin and VEGF of GBC-SD cells were significantly down-regulated,while protein expressions of p21 and E-cadherin,and Bax/Bcl-2 ratio were significantly up-regulated in 5 mmol/L Bupivacaine group(P<0.05).Conclusion Bupivacaine may inhibit the proliferation,invasion and microtubule formation of GBC-SD cells in patients with gallbladder cancer.
