摘 要:目的:探索β2肾上腺素受体阻断剂ICI 118,551逆转颞下颌关节骨关节炎(TMJ-OA)大鼠髁突软骨下骨吸收的效果。方法:建立磨牙咬合紊乱致大鼠TMJ-OA模型(实验组)及其操作对照,实验开始后每周于大鼠腹腔注射生理盐水(溶剂)或ICI 118,551(20μg/g体重)1次,8周后处死大鼠,采用micro-CT检测髁突软骨下骨密度及骨小梁结构的差异,TRAP染色观察软骨下骨破骨细胞活化情况,采用实时定量PCR检测软骨下骨破骨因子基因表达情况。结果:溶剂注射实验组大鼠髁突软骨下骨丢失显著,其骨密度、骨体积分数、骨小梁厚度及数量均较其对照组显著降低,而其骨小梁间隙、破骨细胞数目及面积、破骨相关因子TRAP、组织蛋白酶K及RANKL基因表达则显著增加(P<0.05);ICI 118,551可显著逆转实验组髁突软骨下骨丢失以及破骨活动增强,表现为其骨密度、骨体积分数及骨小梁厚度均较溶剂实验组增高,而骨小梁间隙、破骨细胞数目及面积、破骨因子基因表达则较溶剂实验组降低(P<0.05)。结论:阻断β2肾上腺素受体信号可逆转异常咬合所导致的TMJ-OA髁突软骨下骨丢失。Objective:To explore the effect of ICI 118,551(ICI,selective blocker of β2-adrenoceptor)on reversing subchondral bone resorption in temporomandibular joint osteoarthritis(TMJ-OA)rats.Methods:TMJ-OA rat model was established by experimentally abnormal molars occlusion(experimental group),and their sham controls(control group)were also established.Peritoneal injection of normal saline(vehicle)or ICI(20 μg/g)were performed from beginning of the experiment(once/per week),and all rats were killed after 8 weeks.Micro-CT was used to detect the bone mineral density(BMD)and microstructures of the condylar subchondral bone.TRAP staining was used to observe the activation osteoclasts in condylar subchondral, and real-time PCR was used to detect the gene expression osteoclastic factors.Results:Compared with the control group, obvious condylar subchondral bone loss was observed in the rats from experimental group, characterized as decreased BMD,bone volume fraction(BV/TV)and trabecular thickness(Tb.Th)of subchondral bone, but increased trabecular space(Tb.Sp),osteoclast number(OC.N)and percentage area(OC.S/BS),and gene expression of trap, cathepsin K,rankl and rankl/opg ratio(all P<0.05).Intra-articular injection of ICI largely reversed the subcondylar bone loss and osteoclastic activation in the experimental rats, evidenced by increased BMD,BV/TV and Tb.Th, but decreased Tb.Sp, OC.N and OC.S/BS,and gene expression of osteoclastic factors, comparing to the vehicle-treated experimental rats(all P<0.05).Conclusion:Blocking β2-adrenoceptor signal could reverse the condylar subchondral bone loss in TMJ-OA rat model induced by abnormal molar occlusion.
