摘 要:目的分析表没食子儿茶素没食子酸酯(EGCG)对裸鼠卵巢癌移植瘤生长及血管内皮生长因子(VEGF)和增殖细胞核抗原(PCNA)表达的影响。方法于裸鼠右侧肩胛背部皮下接种0.2 mL SKOV3肿瘤细胞悬液建立人卵巢癌裸鼠皮下移植瘤模型。将建模成功的35只裸鼠按随机数字表法分为低剂量组、中剂量组、高剂量组、对照组和紫杉醇组,每组7只。低剂量组、中剂量组和高剂量组裸鼠每日分别经腹腔注射0.2 mL EGCG(10、30、50 mg/kg);紫杉醇组裸鼠每3 d经腹腔注射0.2 mL紫杉醇(5 mg/kg);对照组裸鼠每3日经腹腔注射0.2 mL 0.9%氯化钠溶液;连续给药21 d。给药期间,于规定时间对裸鼠体重、移植瘤体积、肿瘤抑制率、脏器指数、微血管密度(MVD)、肝组织形态、移植瘤组织中VEGF和PCNA mRNA表达水平及蛋白表达水平进行检测。结果给药21 d后,EGCG各剂量组和紫杉醇组裸鼠肿瘤体积增长速度均明显慢于对照组,肿瘤抑制率均明显高于对照组,差异有统计学意义(P<0.05)。与对照组相比,EGCG各剂量组裸鼠的瘤重、胸腺指数和脾脏指数均显著升高,MVD和肿瘤组织中VEGF和PCNA mRNA表达水平及蛋白表达水平均明显降低,差异均有统计学意义(P<0.05);且随着EGCG给药浓度增大,以上指标的增大或降低幅度增大。EGCG各剂量组裸鼠体重、肝脏指数及肝脏组织形态无显著变化。结论EGCG对SKOV3卵巢癌移植瘤的生长具有明显的抑制作用,其作用机制可能与抑制VEGF、PCNA的表达密切相关。Objective To analyze the effect of epigallocatechin gallate on the growth and expression of VEGF and PCNA of ovarian cancer transplanted tumor in nude mice.Methods Inoculate 0.2 mL SKOV3 tumor cell suspension subcutaneously on the back of the right scapula of nude mice to establish a subcutaneous transplantation tumor model of human ovarian cancer in nude mice.The 35 nude mice successfully modeled were randomly divided into low-dose group,middle-dose group,high-dose group,control group and paclitaxel group according to the random number table method,with 7 mice in each group.Nude mice in the low-dose group,middle-dose group and high-dose group were injected intraperitoneally with 0.2 mL EGCG(10,30,50 mg/kg)daily;nude mice in the paclitaxel group were injected intraperitoneally with 0.2 mL paclitaxel every 3 days(5 mg/kg);control group nude mice were injected intraperitoneally with 0.2 mL of normal saline every 3 days;continuous administration for 21 days.During the administration period,nude mice body weight,transplanted tumor volume,tumor inhibition rate,organ index,microvessel density(MVD),liver tissue morphology,VEGF and PCNA mRNA expression levels and protein expression levels in transplanted tumor tissues were detected at the specified time.Results After 21 days of administration,the tumor volume growth rate of nude mice in each dose of EGCG and paclitaxel group was significantly slower than that in the control group,and the tumor inhibition rate of nude mice was significantly higher than that in the control group,the differences were statistically significant(P<0.05).Compared with the control group,the tumor weight,thymus index and spleen index of nude mice in each dose group of EGCG were significantly increased(P<0.05),and the microvessel density and the expression level of VEGF and PCNA mRNA and protein expression in the tumor group were significantly decreased(P<0.05);And with the increase of the EGCG concentration,the increase or decrease of the above indicators increases.There were no significant chang
