摘 要:目的研究骨肉瘤中miR-200c表达水平与病理特征、病情转归的相关性并探究其潜在靶基因。方法回顾性选择2016年4月至2018年12月期间在华中科技大学同济医学院附属协和医院诊断为骨肉瘤的52例患者。观察骨肉瘤组织与瘤旁组织中miR-200c表达水平的变化;比较不同病理特征骨肉瘤中miR-200c表达水平;随访生存时间、绘制K-M曲线并进行Long-rank检验,分析不同miR-200c表达水平骨肉瘤患者的生存情况;分析骨肉瘤中AKT通路基因的表达及其与miR-200c的相关性。结果骨肉瘤组织中miR-200c的表达水平(0.47±0.09)明显低于瘤旁组织(1.06±0.20),差异有统计学意义(P<0.05)。EnnekingⅡb-Ⅲ期、有淋巴结转移的骨肉瘤组织中miR-200c的表达水平明显低于EnnekingⅠ-Ⅱa期、无淋巴结转移的骨肉瘤组织,差异有统计学意义(P<0.05)。骨肉瘤中miR-200c低表达患者的整体生存情况较miR-200c高表达患者恶化。骨肉瘤组织中蛋白激酶B(AKT)、哺乳动物雷帕霉素白蛋白(mTOR)的表达水平分别为1.88±0.31、1.73±0.30,明显高于瘤旁组织(1.04±0.18、1.02±0.19),差异均有统计学意义(P<0.05)。AKT、mTOR与miR-200c的表达水平呈负相关(r=-0.315,-0.228,P<0.05)。结论骨肉瘤中miR-200c呈低表达趋势并且能够促进肿瘤病理进程、影响远期生存,靶向AKT/mTOR是其可能机制。Objective To study the correlation between the expression level of miR-200c in osteosarcoma and the pathological characteristics,prognosis and to explore its potential target genes.Methods Fifty two patients diagnosed as osteosarcoma in Union Hospital Affiliated to Tongji Medical College,Huazhong University of Science and Technology from April 2016 to December 2018 were retrospectively selected.The level of miR-200c expression in osteosarcoma tissue and adjacent tissues was observed.The level of miR-200c expression in osteosarcoma with different pathological characteristics was compared.Survival time was followed up,K-M curve was drawn and Long-rank test was performed,and the survival of osteosarcoma patients with different miR-200c expression levels was analyzed.The expression of AKT pathway genes in osteosarcoma and its correlation with miR-200c were analyzed the expression of AKT pathway genes in osteosarcoma and its correlation with miR-200c were analyzed.Results The level of miR-200c expression in osteosarcoma tissues was 0.47±0.09,which was significantly lower than that in adjacent tissues(1.06±0.20),the difference was statistically significant(P<0.05).The levels of miR-200c expression in EnnekingⅡb-Ⅲand lymph node metastasis osteosarcoma tissues were significantly lower than those in EnnekingⅠ-Ⅱa and non-lymph node metastasis osteosarcoma tissues,the differences were statistically significant(P<0.05).The overall survival of patients with low expression of miR-200c in osteosarcoma was worse than that of patients with high expression of miR-200c.The levels of protein kinase B(AKT)and mammalian target of rapamycin(mTOR)expression were 1.88±0.31,1.73±0.30 in osteosarcoma tissues,which were significantly higher than those in adjacent tissues(1.04±0.18,1.02±0.19),the differences were statistically significant(P<0.05),and negatively correlated with the levels of miR-200c expression(r=-0.315,-0.228,P<0.05).Conclusion The expression of miR-200c in osteosarcoma tends to decrease and it can promote the
