摘  要:目的观察知葛通脉颗粒对糖尿病动脉粥样硬化(AS)大鼠斑块组织炎性因子蛋白表达水平及对胸主动脉内中膜厚度变化的影响。方法采用随机对照法将46只无特定病原体(SPF)级Wistar雄性大鼠分为对照组、模型组、中药组和西药组。对照组给予基础饲料喂养,其余各组采用亚致病剂量链脲佐菌素(STZ)注射+主动脉内膜球囊拉伤术加高脂饲料喂养构建糖尿病AS模型。中药组给予知葛通脉颗粒,西药组给予瑞舒伐他汀片。给药6周后,胸主动脉剪取粥样斑块留作标本,检测核转录因子-κB(NF-κB)、肿瘤坏死因子-α(TNF-α)、基质金属蛋白酶-9(MMP-9)、血管细胞黏附分子-1(VCAM-1)、单核细胞趋化因子-1(MCP-1)、细胞间黏附分子-1(ICAM-1)、基质金属蛋白酶组织抑制因子(TIMP)蛋白表达水平及胸主动脉内中膜厚度的变化。结果对照组、中药组、西药组与模型组比较NF-κB、TNF-α、MMP-9、VCAM-1、ICAM-1、MCP-1、TIMP蛋白表达水平明显下降(P<0.05);中药组NF-κB、TNF-α、MMP-9、VCAM-1、ICAM-1、MCP-1蛋白表达水平与西药组比较,差异均无统计学意义(P>0.05);西药组TIMP蛋白表达水平比中药组下降更明显(P<0.05);对照组、中药组、西药组与模型组相比较,胸主动脉内中膜厚度明显减小(P<0.05);中药组胸主动脉内中膜厚度与西药组比较,差异无统计学意义(P>0.05)。结论知葛通脉颗粒可以明显改善糖尿病AS大鼠斑块组织NF-κB、TNF-α、MMP-9、VCAM-1、ICAM-1、MCP-1、TIMP的蛋白表达水平,改善胸主动脉内中膜厚度,调整内皮功能。Objective To observe the effect of Zhige Tongmai granules on the protein expression of tissue inflammatory factor and the thickness of the inner middle membrane of thoracic aorta in diabetic AS rats.Methods Fourty-six SPF wistar male rats were randomly divided into four groups:blank group,model group,traditional Chinese medicine(TCM)group,and western medicine group.The rats in blank group were fed with basic feed.The other groups were treated with subpathogenic dose of streptozotocin injection.Aortic intima balloon aspiration and high-fat diet were used to establish diabetic AS model.The rats in TCM group were given Zhige Tongmai granules,the rats in western medicine group were given rosuvastatin tablets.Four weeks after administration,plaque was cut from thoracic aorta as specimen for detection the protein expression levels of NF-κB,TNF-α,MMP-9,VCAM-1,MCP-1,ICAM-1,TIMP and the thickness of the inner middle membrane of thoracic aorta were detected.Results Compared with the model group,the protein expression levels of NF-κB,TNF-α,MMP-9,VCAM-1,ICAM-1,MCP-1,and TIMP in blank group,TCM group and western medicine group were significantly decreased(P<0.05).There were no significantly differences between TCM group and western medicine group in the protein expression levels of NF-κB,TNF-α,MMP-9,VCAM-1,ICAM-1,and MCP-1(P>0.05),the protein expression levels of TIMP decreased more obviously in the western medicine group than that of the TCM group(P<0.05).Compared with the model group,the thickness of the inner middle membrane of thoracic aorta in blank group,TCM group and western medicine group were significantly decreased(P<0.05).There were no significantly differences between TCM group and western medicine group in the thickness of the inner middle membrane of thoracic aorta(P>0.05).Conclusion Zhige Tongmai granules can significantly decrease the protein expression levels of NF-κB,TNF-α,MMP-9,VCAM-1,ICAM-1,MCP-1,and TIMP in diabetic AS rats,decreased the thickness of the inner middle membrane of thoracic aorta,reg
