摘  要:目的研究肠上皮细胞中多配体蛋白聚糖1(SDC-1)、富亮氨酸α-2糖蛋白1(LRG1)的表达与溃疡性结肠炎(UC)患者屏障功能损伤的相关性。方法该研究采用前瞻性研究方法,选择2018年1月至2020年5月在该院进行诊断及治疗的UC患者120例作为观察组,根据简化UC活动指数(CDAI)分析,缓解期患者56例,活动期患者64例,另选取同期该院治疗的肠息肉患者120例作为对照组,采用免疫组化法对患者的SDC-1、LRG1水平进行检测,比较观察组与对照组,不同严重程度UC患者的肠上皮细胞中SDC-1、LRG1及肠道黏膜的肠道屏障指标,分析肠上皮细胞中SDC-1、LRG1的表达与溃疡性结肠炎患者屏障功能损伤的相关性。结果观察组患者的SDC-1、LRG1水平显著低于对照组,差异有统计学意义(P<0.05);缓解组患者的SDC-1、LRG1水平显著高于进展组,差异有统计学意义(P<0.05);观察组患者的肠道屏障指标一氧化氮(NO)、半乳糖凝集素3(galectin-3)、密封蛋白(OCLN)、galectin-1、紧密连接蛋白1(ZO-1)及扣带蛋白(cingulin)水平显著低于对照组,差异有统计学意义(P<0.05);缓解组患者的一氧化氮(NO)、galectin-3、OCLN、galectin-1、ZO-1及cingulin水平显著低于对照组,差异有统计学意义(P<0.05);相关性分析显示,患者的SDC-1、LRG1水平与NO、galectin-3、OCLN、galectin-1、ZO-1及cingulin水平呈正相关。结论肠上皮细胞中SDC-1、LRG1的表达与溃疡性患者屏障功能损伤显著相关性,可作为临床诊断的重要依据。Objective To study the relationship between the expression of syndecan-1(SDC-1),leucine-rich-alpha-2-glycoprotein1(LRG1)in intestinal epithelial cells and barrier function injury in ulcerative colitis(UC)patients.Methods This study used a prospective research method.A total of 120 patients with UC diagnosed and treated in the hospital from January 2018 to May 2020 were selected as the observation group.According to the simplified UC activity index(CDAI)analysis,56 patients were in remission stage,64 patients were in active stage.Another 120 patients with intestinal polyps treated in the hospital during the same period were selected as the control group.The levels of SDC-1 and LRG1 in patients were detected by immunohistochemistry.The levels of SDC-1 and LRG1 in intestinal epithelial cells of patients with ulcerative colitis of different severity were compared between the observation group and the control group,as well as the intestinal barrier indexes of intestinal mucosa.The correlation between the expression of SDC-1 and LRG1 in intestinal epithelial cells and the barrier function damage of patients with ulcerative colitis was analyzed.Results The levels of SDC-1 and LRG1 in the observation group were significantly lower than those in the control group,and the differences were statistically significant(P<0.05).The levels of SDC-1 and LRG1 in the remission group were significantly higher than those in the progressive group,and the differences were statistically significant(P<0.05).The intestinal barrier indexes of nitric oxide(NO),galectin-3,occludin(OCLN),galectin-1,zonula occludens 1(ZO-1)and cingulin in the observation group were significantly higher than those in the control group,and the differences were statistically significant(P<0.05).NO,galectin-3,OCLN,galectin-1,ZO-1 and cingulin in the remission group were significantly higher than those in the progressive group,and the differences were statistically significant(P<0.05).The levels of SDC-1 and LRG1 were positively correlated with the levels of NO,gale
