摘 要:目的探究白细胞介素-17A(IL-17A)、IL-17F和IL-23R基因多态性与结直肠癌易感性的关系。方法收集该院2016年5月至2019年5月确诊的140例结直肠癌患者(实验组)和100例进行健康体检的体检健康者(对照组)资料,抽取外周血样血细胞DNA后,利用聚合酶联反应-连接酶反应检测技术(PCR-LDR)分型技术检测样本中IL-17A(rs2275913)位点、IL-17F(rs763780)位点和IL-23R(rs10889677)位点基因多态性。结果两组IL-17A(rs2275913)TT、TC和CC分布情况差异无统计学意义(P>0.05)。实验组IL-17F(rs763780)TT、TC和CC分布情况与对照组比较差异有统计学意义(P<0.05),但其等位基因分布差异无统计学意义(P>0.05)。实验组IL-23R(rs10889677)TT、TC和CC分布情况与对照组比较差异有统计学意义(P<0.05),但其等位基因分布比较差异无统计学意义(P>0.05)。IL-17F(rs763780)单核苷酸基因频率与肿瘤分期相关,纯合子TT在中重度患者中分布明显少于轻度患者,差异有统计学意义(P<0.05),且肿瘤位于升结肠的纯合子TT基因频率明显异于其他部位,差异有统计学意义(P<0.05);IL-23R(rs10889677)位点单核苷酸基因频率与肿瘤分期及肿瘤部位差异无统计学意义(P>0.05)。Logistic回归分析显示,IL-17F(rs763780)Ⅲ、Ⅳ期患者中TT基因型明显异于其他(CT+CC)基因型,差异有统计学意义(P<0.05);肿瘤位于升结肠患者中IL-23R(rs10889677)CC基因型明显异于其他(TT+TC)基因型,差异有统计学意义(P<0.05)。结论IL-17F(rs763780)和IL-23R(rs10889677)可能影响结直肠癌患者的易感性,其多核苷酸多态性分别与患者肿瘤分期、肿瘤部位相关。Objective To explore the relationship between interleukin(IL)-17A,IL-17F and IL-23R gene polymorphisms and colorectal cancer susceptibility.Methods A total of 140 patients with colorectal cancer(experiment group)and 100 healthy patients(control group)from May 2015 to May 2019 were collected.After extracting DNA from peripheral blood samples,the gene polymorphism of IL-17A rs2275913 locus,IL-17F rs763780 locus and IL-23R rs10889677 locus in the sample were detected by polymerase chain reaction ligase reaction(PCR-LDR)typing.Results There was no significant difference in the distribution of IL-17A(rs2275913)TT,TC and CC between the two groups(P>0.05).There were significant differences in the distribution of TT,TC and CC of IL-17F(rs763780)between the experience group and the control group(P<0.05),but there was no significant difference in the distribution of alleles(P>0.05).There were significant differences in the distribution of TT,TC and CC of IL-23R(rs10889677)between the experience group and the control group(P<0.05),but there was no significant difference in the distribution of alleles(P>0.05).The frequency of IL-17F(rs763780)single nucleotide gene was correlated with tumor stage,the distribution of homozygous TT in moderate to severe patients was significantly less than that in mild patients,and the difference was statistically significant(P<0.05).The frequency of homozygous TT gene in ascending colon was significantly different from that in other parts(P<0.05).The frequency of IL-23R(rs10889677)single nucleotide gene was not significantly different from that in tumor stage and tumor location(P>0.05).Logistic regression analysis showed that TT genotype was significantly different from other(CT+CC)genotypes in IL-17F(rs763780)stageⅢandⅣpatients,and the difference was statistically significant(P<0.05).IL-23R(rs10889677)CC genotype in patients with tumor located in ascending colon was significantly different from other(TT+TC)genotypes(P<0.05).Conclusion IL-17F(rs763780)and IL-23R(rs10889677)may affect the s
