摘 要:【目的】研究IBA对费约果扦插生根的影响,探究插穗生根进程中形态、内源激素水平、氧化酶活性和营养物质的含量变化,初步揭示费约果插穗生根机制。【方法】以不同浓度IBA处理费约果半木质化插穗,记录生根情况,根据形态变化划分生根阶段。插后每隔10 d取样,检测IBA对插穗内源激素水平、氧化酶活性及营养物质含量的影响。【结果】1)IBA处理促进费约果生根,其中1000 mg/L IBA处理下费约果生根率达69.13%,优于其他处理。费约果生根类型为诱导生根型,生根阶段分别为愈伤组织期(0~20 d)、根原基诱导期(20~40 d)、不定根表达期(40~60 d)、不定根伸长期(60 d后)。2)IBA调节费约果插穗内源激素变化,插穗IAA含量与IAA/ZR值先上升后下降,在30 d达峰值。ZR与ABA含量呈下降趋势,IAA/ABA值在30、50 d出现两个峰值。氧化酶活性变化规律明显,IAAO、POD、PPO活性均先升高再降低,IBA处理峰值比CK提前10 d。营养物质含量变化中,可溶糖含量变化明显,可溶蛋白含量先上升后下降,(可溶糖+淀粉)/可溶蛋白与淀粉含量先下降后上升。【结论】1000 mg/L IBA促进费约果生根的效果优于其他处理。高浓度IAA及低浓度ABA、ZR利于根系诱导。IBA刺激插穗合成IAA,加快ABA与ZR消耗,提高IAA/ABA、IAA/ZR值。低的IAAO活性及高的POD、PPO活性共同调节插穗IAA水平来促进生根。IBA加快营养物质代谢速度与强度,高浓度可溶性蛋白利于根原基诱导,淀粉及时水解为可溶性糖整个生根过程提供能量。【Objective】To study the effect of IBA on of Feijoa cutting rooting,exploring the changes of morphology,hormone level,oxidase activity and nutrient content in the process of rooting,and reveal the rooting mechanism of Feijoa cuttings.【Method】Different concentrations of IBA were used to treat the semi-lignified cuttings of Feijoa,record the rooting situation,and divide the rooting stage according to the morphological changes.Sampling every 10 days after cutting to detect the effect of IBA on endogenous hormone levels,oxidase activity and nutrient content of cuttings.【Result】1)IBA treatment promote the rooting of Feijoa,and the rooting rate of Feijoa is 69.13%under 1000 mg/L IBA treatment,better than other treatments.The rooting type of Feijoa is induced rooting.The rooting stages are callus period(0-20 d),root primordium induction period(20-40 d),adventitious root expression period(40-60 d),adventitious root elongation period(after 60 d).2)IBA adjust the endogenous hormone changes of Feijoa cuttings.The IAA content and IAA/ZR value of cuttings show the tendency rising up at the begining and declining in late,reaching the peak at 30 days.The contents of ZR and ABA show a downward trend.The IAA/ABA value show two peaks at 30 d and 50 d.The activity of oxidase changes obviously.The activities of IAAO,POD and PPO all increase first and then decrease.The peak value of IBA treatment is 10 days earlier than that of CK treatment.In the nutrient content changes,The soluble sugar content has changed significantly,the soluble protein content first increase and then decrease,(soluble sugar+starch)/soluble protein and starch content decrease and then increase.【Conclusion】The effect of 1000 mg/L IBA on promoting the rooting of Feijoa is better than other concentrations.High concentration of IAA,IAA/ABA,IAA/ZR and low concentration of ABA and ZR were beneficial to root formation.IBA stimulates cuttings to synthesize more endogenous hormone IAA,promotes the consumption of ABA and ZR,and increases the ratio of IAA/A
