摘  要:为研究北艾根际和非根际土壤微生物群落结构和多样性,利用Illumina高通量测序技术对汤阴北艾根际和非根际的土壤样品进行对比分析。结果表明,北艾根际土壤真菌、细菌序列数分别比非根际土壤多16 813条和3 972条,北艾根际土壤微生物的Shannon指数、Chao1指数、Ace指数都大于非根际土壤。从真菌组成分析,2种土壤真菌优势门均为子囊菌门(Ascomycota),主要优势属种类存在较大差异;北艾根际土壤中的优势属是unclassified_Xylariales和柄孢壳菌属(Podospora),非根际土壤中的优势属是unclassified_Dothideomycetes和镰刀菌属(Fusarium);此外,北艾根际土壤中具有特有的球囊菌门(Glomeromycota),而非根际土壤中发现有白蘑科(Tricholomataceae)。从细菌组成分析,2种土壤细菌的优势门均为变形菌门(Proteobacteria),优势属均为鞘氨醇单胞菌属(Sphingomonas)。主成分分析表明,土壤微生物多样性及其丰富度受北艾的种植作用影响较大,北艾根际土壤微生物群落结构与非根际土壤存在一定差异,其多样性和丰富度大于非根际土壤。In order to study the microbial community structure and diversity in the rhizosphere and non-rhizosphere soils of Artemisia vulgaris,the related soil samples were compared and analyzed by Illumina Next-generation sequencing(NGS)technology.The results showed that there were 16813 and 3972 more fungal and bacteria sequences in the rhizosphere soil than non-rhizosphere soil,respectively.The Shannon index,Chao1 index,and Ace index of rhizosphere soil microorganisms in Artemisia vulgaris were greater than those of non-rhizosphere soil.According to the fungal composition analysis,the dominant fungi phyla of two soil fungi were Ascomycota,but there were significant differences in the dominant genera.The dominant genera in the rhizosphere soil were Unclassified Xylariales and Podospora.The dominant genera in non-rhizosphere soils were Unclassified Dothideomycetes and Fusarium.In addition,the unique Glomeromycota was found in the rhizosphere soil of Artemisia vulgaris,while the Tricholomataceae fungi was found in the non-rhizosphere soil.The analysis of bacterial composition showed that the dominant soil phyla and the dominant genus in both the rhizosphere and non-rhizosphere soils were Proteobacteria and Sphingomonas,respectively.Principal component analysis showed that the soil microbial diversity and its abundance were greatly affected by the planting effect of Artemisia vulgaris.The microbial community structure in the rhizosphere soil of Artemisia vulgaris was different from that in the non-rhizosphere soil,and the microbial community structure diversity and the abundance of rhizosphere soil were more abundant than those of non-rhizosphere soil.
