摘 要:目的:研究隔日禁食结合济饥辟谷方对非酒精性脂肪性肝病(NAFLD)大鼠肝脏氧化应激及胰岛素受体底物(IRS)2的影响。方法:44只雄性SD大鼠随机分为正常对照组(n=12)及高脂饲料组(n=32),于12周末从各组随机抽取2只大鼠处死。建模成功后,将高脂饲料组分成模型组、隔日禁食组、结合济饥辟谷方组,每组10只。于16周末采血检测总胆固醇(TC)、甘油三酯(TG)、丙氨酸氨基转移酶(ALT)和天门冬氨酸氨基转移酶(AST),取肝组织作油红O、TUNEL和HE染色,并测定丙二醛(MDA)含量、超氧化物歧化酶(SOD)和谷胱甘肽(GSH)活性、IRS 2 mRNA表达。结果:与正常对照组比较,模型组肝脏脂质沉积、炎性细胞浸润及肝细胞损伤(如气球样变、凋亡及坏死),肝脏MDA含量升高、SOD和GSH活性降低、IRS 2 mRNA表达下调,血清TC、TG、ALT和AST水平升高(P<0.05)。与模型组比较,隔日禁食组及结合济饥辟谷方组均能减少肝脏脂质沉积、炎性细胞浸润及肝细胞损伤,促进肝脏及血清各指标的恢复(P<0.05),并且结合济饥辟谷方组的效果优于隔日禁食组(P<0.05)。结论:隔日禁食结合济饥辟谷方可显著减轻NAFLD大鼠肝脏氧化应激及胰岛素抵抗,改善肝细胞损伤和脂质沉积,其发挥作用的机制与提高抗氧化酶活性及IRS 2 mRNA表达有关。Objective:To study the effect of ADF combined with JJBGP on the liver oxidative stress and IRS 2 in NAFLD rats.Methods:Forty-four healthy male SD rats were randomly divided into the normal control group and the high-fat diet group,and 2 rats were randomly selected from each group at the end of the 12th week and sacrificed.After successful modeling,the high-fat group was divided into model group,ADF with or without JJBGP group.Blood samples were collected at the end of the 16th week to detect total cholesterol(TC),triglycerides(TG),alanine aminotransferase(ALT)and aspartate aminotransferase(AST).Liver tissues were stained with Oil Red O,TUNEL and HE,respectively,and were used to detect superoxide dismutase(SOD),glutathione(GSH),malondialdehyde(MDA)and IRS 2 mRNA expression.Results:Compared with the normal control group,the model group showed,liver lipid deposition,inflammatory cell infiltration and liver cell damage(such as ballooning,apoptosis and necrosis),increased liver MDA content,decreased SOD and GSH activities,IRS 2 reduced mRNA expression,TC,TG,ALT and AST increased in serum(P<0.05).Compared with the model group,ADF with or without JJBGP could reduce liver lipid deposition,inflammatory cell infiltration and liver cell damage,and promote the recovery of liver and serum indicators(P<0.05).In addition,the combined treatment was better than fasting alone(P<0.05).Conclusion:ADF combined JJBGP could significantly improve liver oxidative stress and insulin resistance,along with liver cell damage and lipid deposition in NAFLD rats.The mechanism of its action was related to the increase of antioxidant enzyme activity and IRS 2 mRNA expression.
