摘  要:目的:观察三黄柴术方对急性肝内胆汁淤积性肝损伤的防治作用及对肝组织水通道蛋白8/9表达的影响。方法:SD雄性大鼠随机分成正常组、模型组、复方高剂量组、复方低剂量组,每组8只。复方高、低剂量组大鼠从实验第1天起分别灌服20 g/(kg·d)、10 g/(kg·d)的三黄柴术方水提液,每天1次,连续4天;正常组和模型组大鼠每天灌服等容量的生理盐水。实验第3天上午正常组大鼠灌服1 ml/kg麻油,其他3组均1次性灌服1 ml/kg 4%萘异硫氰酸酯(ANIT)麻油溶液。实验第5天上午采集标本,采用全自动生化仪检测实验大鼠血清肝功能指标,HE染色观察实验大鼠肝组织病理学变化,qRT-PCR和Western Blot技术检测实验大鼠肝组织AQP8 mRNA、AQP9 mRNA及其蛋白表达水平。结果:模型组大鼠血清ALT、AST、ALP、TBil、TBA水平较正常组明显升高(P<0.01);复方高、低剂量组大鼠血清ALT、AST水平较模型组明显降低(P<0.01)。模型组大鼠肝组织出现肝细胞脂肪变,汇管区、小叶周边出现小的胆管增生和少量胶原沉积,有少量炎性细胞浸润;复方高、低剂量组大鼠肝组织病理变化程度明显减轻。模型组大鼠肝组织AQP8、AQP9 mRNA和蛋白表达均较正常组明显下调(P<0.01),复方高剂量组大鼠肝组织AQP8、AQP9 mRNA和蛋白表达均较模型组显著升高(P<0.01或P<0.05),复方低剂量组大鼠肝脏AQP8 mRNA、AQP9 mRNA和AQP 9蛋白表达较模型组显著增高(P<0.05)。结论:ANIT诱导的急性肝内胆汁淤积大鼠肝组织AQP8、AQP9基因及蛋白质表达水平均下降,三黄柴术方能够上调实验大鼠肝组织AQP8/9表达,这可能是其改善大鼠肝内胆汁淤积性肝细胞损伤的作用机制之一。Objective:To observe the effect of Sanhuang Chaishu Decoction on the prevention and treatment of acute intrahepatic cholestasis liver injury and to explore the possible molecular mechanism from the water channel protein(Aquaporins,AQPs).Methods:SD rats were randomly divided into 4 groups,including normal group,model group,Sanhuang Chaishu Decoction groups(low and high dose),8 rats in each group.Compound high and low dose group from the first day orally 20 g/(kg·d),10 g/(kg·d)of Sanhuang Chaishu Decoction,1 times a day for 4 days,normal group and model group with the equal volume of NS gavage;the normal group was fed with 1ml/kg sesame oil in the morning of the third day,while the model group,compound high and low dose group were administered 4% of ANIT(made up of sesame oil,1 ml/kg)once;Rats in each group were treated in the morning on the fifth day of the experiment.Detecting serum liver function by automatic biochemical analyzer.HE staining,observe pathological changes of liver tissue.RT-PCR and Western Blot techniques were used to detect the mRNA and protein expression of AQP8/9.Results:The levels of serum ALT,AST,ALP,TBil and TBA in the model group were significantly higher than those in the normal group(P<0.01);The levels of serum ALT and AST in the high and low dose groups were significantly lower than those in the model group(P<0.01).In the model group,hepatocyte steatosis,small bile duct hyperplasia,a small amount of collagen deposition and a small amount of inflammatory cell infiltration appeared in the portal area and around the lobules;The pathological changes of liver tissue in high and low dose groups were significantly reduced.The expression of AQP8,AQP9 mRNA and protein in liver tissue of model group was significantly lower than that of normal group(P<0.01),the expression of AQP8,AQP9 mRNA and protein in liver tissue of compound high-dose group was significantly higher than that of model group(P<0.01 or P<0.05),and the expression of AQP8 mRNA,AQP9 mRNA and AQP9 protein in liver of compound low-do
