摘 要:目的观察微小RNA(miRNA,miR)-1277-5p过表达对卵巢癌SKOV-3细胞增殖和凋亡的影响并探讨其机制。方法将对数生长期的SKOV-3细胞分为NC组和miR-1277-5p组,分别转染对照模拟物和miR-1277-5p模拟物。实时荧光定量PCR(qRT-PCR)检测各组SKOV-3细胞miR-1277-5p表达水平。集落形成实验检测各组SKOV-3细胞增殖。流式细胞术检测SKOV-3细胞凋亡变化。RNAhybrid预测miR-1277-5p的靶基因。qRT-PCR和蛋白质印迹(Westernblot)检测靶基因mRNA和蛋白的表达。结果NC组和miR-1277-5p组SKOV-3细胞中miR-1277-5p的表达分别为(1.22±0.42)和(13.22±1.42),NC组明显少于miR-1277-5p组(P<0.01)。与NC组比较,miR-1277-5p组集落的数量较少(P<0.01),细胞凋亡率明显较高(P<0.01)。miR-1277-5p的靶基因可能是PHD锌指蛋白8(PHF8)。与NC组比较,miR-1277-5p组SKOV-3细胞中PHF8基因表达明显降低(P<0.01)。结论miR-1277-5p可能通过抑制PHF8基因表达,降低卵巢癌SKOV-3细胞的增殖并促进其凋亡。Objective To observe the effect of microRNA(miRNA,miR)-1277-5p overexpression on the proliferation and apoptosis of ovarian cancer SKOV-3 cells and explore its mechanism.Methods The SKOV-3 cells in logarithmic growth phase were divided into a NC group and a miR-1277-5p group,and the control mimic and miR-1277-5p mimic were transfected respectively.The expression of miR-1277-5p of SKOV-3 cells in each group was detected by qRT-PCR,the proliferation of SKOV-3 cells in each group was detected by the colony formation experiment,and the apoptosis of SKOV-3 cells was detected by the flow cytometry.RNAhybrid was used to predict the target gene of miR-1277-5p,and the expressions of mRNA and protein of target gene were detected by qRT-PCR and Western blot.Results The expressions of miR-1277-5p of SKOV-3 cells in the NC group and miR-1277-5p group were(1.22±0.42)and(13.22±1.42),respectively,which was significantly lower in the NC group than in the miR-1277-5p group(P<0.01).Compared with those in the NC group,the number of colonies in the miR-1277-5p group was fewer(P<0.01),and the apoptosis rate was significantly higher(P<0.01).The target gene of miR-1277-5p might be PHD zinc finger protein 8(PHF8).Compared with that in the NC group,the expression of PHF8 gene of SKOV-3 cells in the miR-1277-5p group was significantly decreased(P<0.01).Conclusion miR-1277-5p may reduce the proliferation of ovarian cancer SKOV-3 cells and promote their apoptosis by inhibiting the expression of PHF8 gene.
