摘  要:为了明确车前草治疗隐匿性肾炎的作用机制,本研究利用网络药理学的方法通过中医药数据库及分析平台和Swiss Target Prediction数据库获得车前草活性成分及对应基因靶点;利用在线人类孟德尔遗传和Genecards数据库获取隐匿性肾炎的基因靶点;在Venny2.1.0平台对车前草靶点及隐匿性肾炎靶点进行重叠;采用String数据库和Cytoscape3.7.1软件对交集基因进行蛋白互作网络分析;再利用R语言对交集基因进行基因本体和京都基因与基因组百科全书富集分析;最后对重要活性成分进行分子对接预测。结果表明车前草可能通过蛋白酪氨酸激酶等生物功能过程,参与PI3K-Akt信号通路达到治疗隐匿性肾炎的目的。分子对接结果显示了重要化合物对AKT1、TP53、VEGFA具有很好的亲和能力。明确了车前草通过调控多种抗炎症基因,参与多条信号通路达到治疗隐匿性肾炎的目的,为进一步探究其作用机制提供一定理论基础。This study uses network pharmacology to obtain the active ingredients and corresponding gene targets of plantain via the Traditional Chinese Medicine Database and Analysis Platform and the Swiss Target Prediction database to clarify the mechanism of plantain herb in the treatment of latent nephritis.The gene targets of latent nephritis were firstly obtained using Online Mendelian Inheritance in Man and Genecards database,and the plantain target and latent nephritis target were overlapped on the Venny 2.1.0 platform.Protein interaction network on the intersection genes were then analyzed using the String database and Cytoscape 3.7.1 software,Gene Ontology and Kyoto Encyclopedia of Genes and Genomes enrichment analysis on the intersection genes was performed by R language.Finally,molecular docking for important active ingredients was predicted.The results show that plantain herb may participate in the PI3K-Akt signaling pathway through protein tyrosine kinase and other biological functional processes to achieve the purpose of treating occult nephritis.The molecular docking results show that important compounds have a good affinity to RAC-alpha serine/threonine-protein kinase,cellular tumor antigen p53,and vascular endothelial growth factor A.It demonstrates that plantain can regulate multiple anti-inflammatory genes and participate in multiple signal pathways to achieve the treatment of latent nephritis,which provides a theoretical basis for further exploration of its mechanism of action.
