摘　　要：目的探讨长链非编码RNA WT1-AS预测胃癌新辅助化疗疗效的临床价值。方法回顾性分析63例行新辅助化疗的进展期胃癌临床资料,并检测胃癌组织WT1-AS表达,采用单因素、多因素Logistic回归模型分析临床系数与新辅助化疗疗效的相关性,ROC曲线检测WT1-AS预测化疗疗效的效力;体外检测经不同浓度5-Fu处理的WT1-AS正常表达和过表达胃癌细胞的凋亡情况。结果新辅助治疗有效与否和肿瘤分化程度、TNM分期和WT1-AS表达显著相关(P<0.05),肿瘤分化程度、TNM分期和WT1-AS表达是进展期胃癌新辅助化疗疗效的独立预测因子;WT1-AS促进5-Fu致细胞凋亡(P<0.05)。结论WT1-AS可作为进展期胃癌新辅助疗效的潜在预测因子。Objective To study the clinical value of LncRNA WT1-AS in predicting the efficacy of neoadjuvant chemotherapy for gastric cancer.Methods The clinical data of 63 patients with advanced gastric cancer who underwent neoadjuvant chemotherapy were retrospectively analyzed.The expression of WT1-AS in gastric cancers was detected.The relationship between clinical characteristics and the efficacy of neoadjuvant chemotherapy was analyzed by univariate and multivariate Logistic regression models.The ability of WT1-AS in predicting chemotherapy efficacy was explored by ROC curve.Furthermore,cultured SGC7901 cells were transfected with LncRNA WT1-AS to study the effect of WT1-AS on resistance to 5-Fu chemotherapy.Results The efficacy of neoadjuvant chemotherapy for gastric cancer was significantly correlated with tumor differentiation,TNM stage and WT1-As expression(P<0.05).The degree of tumor differentiation,TNM stage and WT1-As expression were independent predictors for efficacy of neoadjuvant chemotherapy in advanced gastric cancer.In vitro study showed that WT1-AS promoted apoptosis induced by 5-Fu.Conclusion LncRNA WT1-AS may be used as a potential predictor for efficacy of neoadjuvant chemotherapy in advanced gastric cancer.
