摘 要:采用SSR荧光标记和SNP分子标记的KASP技术对豫玉22玉米杂交种子进行基因组DNA提取、引物筛选、基因型分析、纯度鉴定,以期为快速有效鉴定玉米种子纯度提供依据。结果表明:豫玉22的SSR特异性引物为umc2007y4和bnlg161k8,纯度鉴定结果为97.92%;SNP特异性引物为MaSNP64和MaSNP245,纯度鉴定结果为98.44%。因此,SSR和SNP分子标记是鉴定豫玉22玉米杂交种子纯度的最佳方法。Using KASP technology by SSR fluorescent labeling and SNP molecular marker,the genomic DNA of Yuyu 22 maize hybrid seeds was extracted,and the primers were screened,genotypes were analyzed and purity was identified,in order to provide basis for rapid and effective identification of maize seed purity.The results showed that the SSR specific primers of Yuyu 22 were umc2007y4 and bnlg161k8,and the purity was 97.92%.The SNP specific primers were MaSNP64 and MaSNP245,and the purity was 98.44%.Therefore,SSR and SNP molecular markers were the best methods to identify the purity of Yuyu 22 maize hybrid seeds.
