摘 要:目的获得北京市顺义区零售市场生猪肉中小肠结肠炎耶尔森菌的分布及病原特征。方法采集样本,应用细菌增菌培养并同时对增菌液进行荧光PCR预筛查的方法检测小肠结肠炎耶尔森菌。对于鉴定阳性菌株应用传统PCR及多重荧光PCR方法分析5种毒力基因的分布特征。应用肉汤稀释法获得分离菌株的耐药特征并应用脉冲场凝胶电泳(PFGE)进行菌株分子分型。结果70份样本中19份增菌液foxA基因实时荧光PCR筛查阳性,阳性率为27.14%(19/70)。19份foxA筛查阳性标本培养获得小肠结肠炎耶尔森菌。19株菌对头孢唑啉、氨苄西林、头孢西丁、氨苄西林/舒巴坦耐药率分别为97.74%、84.21%、47.39%和42.11%,其携带毒力基因特征均为ail-/ystA-/ystB+/yadA-/virF-。17株菌完成PFGE检测并被分为15种带型。结论针对增菌液进行荧光PCR预筛查,可提高食品样本中小肠结肠炎耶尔森菌分离培养的检出率。菌株毒力基因的荧光PCR组合检测可快速获得分离菌株的毒力特征。本次研究中分离自本地区零售生猪肉中的小肠结肠炎耶尔森菌未发现显著遗传聚集性特征。This study aimed to determine the distribution and characteristics of Yersinia enterocolitica from retail raw pork samples in Shunyi,Beijing.Seventy samples were collected from local markets.Bacterial enrichment isolation and real-time PCR pre-screening for Y.enterocolitica were performed on the collected samples.Antibiotic susceptibility tests and pulsed field gel electrophoresis were performed on the Y.enterocolitica isolates.The presence of toxic genes was analyzed with both conventional and real-time PCR.A total of 19 samples were positive for foxA in the enrichment broth,and 19 Y.enterocolitica isolates were obtained from those 19 samples.The characteristics of the toxic genes(ail-/ystA-/ystB+/yadA-/virF-)of the 19 Y.enterocolitica isolates were identical.The drug resistance rates of the 19 isolates to cefazolin,ampicillin,cefoxitin,and ampicillin/sulbacillin were 97.74%,84.21%,47.39%,and 42.11%,respectively.Seventeen strains were detected by PFGE,and the results were divided into 15 patterns.Real-time PCR pre-screening for enrichment might significantly increase the detection rate of Y.enterocolitica in raw meat samples.No genetic clustering was found among the tested Y.enterocolitica isolates in this study.
