摘 要:目的探讨肌营养不良蛋白结合蛋白1(DTNBP1)基因多态性与中国北方地区汉族人群精神分裂症易感性之间的关联。方法收集精神分裂症汉族患者150例(病例组)和汉族健康志愿者160例(对照组)外周血样本,提取基因组DNA。采用多重高温连接酶检测反应技术(improved multiple ligase detection reaction,iMLDR)对DTNBP1基因的rs909706、rs2619539、rs9370822和rs4236167位点进行基因分型。结果DTNBP1基因的rs909706、rs2619539、rs9370822和rs4236167位点的等位基因、基因型和单倍型频率在病例组和对照组间的分布差异无统计学意义(P>0.05)。按照性别分组rs909706位点的等位基因频率在男性、女性组间的差异具有统计学意义(P=0.000),携带有C等位基因的男性患精神分裂症的风险增高。结论DTNBP1基因的rs909706位点与中国北方地区汉族男性人群精神分裂症的易感性相关。Objective To investigate the association between the polymorphism of dystrobrevin-binding protein 1(DTNBP1)gene and the susceptibility to schizophrenia(SCZ)in northern Han population.Methods Genomic DNA was extracted from the peripheral blood samples of 150 patients of schizophrenia(case group)and 160 healthy Han volunteers(control group).The genotypes of rs909706,rs2619539,rs9370822 and rs4236167 in DTNBP1 gene were detected by the improved multiple ligase detection reaction(iMLDR).There was no significant difference in allele,genotype and haplotype frequencies of rs909706,rs2619539,rs9370822 and rs4236167 in DTNBP1 gene between the case and control group(P>0.05).The allele frequency of rs909706 was significantly different between male and female groups(P=0.000),and men carrying the C allele had an increased risk of schizophrenia.Conclusion The rs909706 polymorphism of DTNBP1 gene is associated with the susceptibility to schizophrenia in northern Chinese Han male population.
