摘  要:目的:探讨太赫磁波(THz)对血管性痴呆(VD)模型大鼠神经行为学及脑组织VEGF/VEGFR2信号通路的影响。方法:采用改良双侧颈总动脉结扎法制备VD模型后,随机分为假手术组、模型组、西药组(尼莫地平,11mg/kg),低、中、高THz组,治疗30d后,Morris水迷宫评价各组大鼠行为学差异,尼氏染色观察海马CA1及皮质区神经锥体细胞变化情况,免疫组化检测脑组织皮质及CA1区中VEGF、VEGFR2蛋白表达。结果:与模型组比较,THz组逃避潜伏期、游泳总路程及首次抵达平台时间均显著缩短(P<0.01,P<0.05),目标象限停留时间显著延长(P<0.05,P<0.01),皮质及CA1区VEGF、VEGFR2的阳性细胞数量显著增加(P<0.05,P<0.01);与西药组比较,中THz组VEGF、VEGFR2阳性表达更强(P<0.05)。结论:THz治疗改善VD模型大鼠学习记忆能力可能与激活脑组织VEGF/VEGFR2信号通路,促血管新生有关。Objective: To investigate the effects of terahertz magnetic wave(THz) on neurobehavioral and VEGF/VEGFR2 signaling pathway in brain tissue of vascular dementia(VD) model rats. Methods: VD models were prepared by vascular occlusion method(2-VO), and they were randomly divided into sham operation group, model group, western medicine group nimodipine, 11 mg/kg, low, medium, and high THz group, treatment for 30 days later, Morris water maze was used to evaluate the behavioral differences of rats in each group. Nissl staining was used to observe the changes of hippocampal CA1 and cortical pyramidal cells, and immunohistochemistry was used to detect the expression of VEGF and VEGFR2 in the cortex and CA1 of brain tissue. Results: Compared with the model group, the escape latency, total swimming distance and first arrival time at the platform in THz group were significantly shorter(P<0.01, P<0.05), the residence time in the target quadrant was significantly prolonged(P<0.05, P<0.01), and the number of VEGF and VEGFR2 positive cells in the cortex and CA1 area was significantly increased(P<0.05, P<0.01). Compared with the western medicine group, the VEGF, VEGFR2 positive expression in the middle THz group was stronger(P<0.05). Conclusion: THz treatment improves the learning and memory ability of VD model rats, which may be related to the activation of the VEGF/VEGFR2 signaling pathway in brain tissue and the promotion of angiogenesis.
