摘 要:为解决珠芽黄魔芋(Amorphophallus bulbifer)产业高质量规模化发展的种苗瓶颈问题,以MS为基本培养基、珠芽黄魔芋块茎为外植体,开展外植体灭菌、愈伤组织诱导、不定芽增殖和生根壮苗试验,筛选珠芽黄魔芋的最佳组培快繁方法。结果表明,最佳外植体灭菌方法为0.3%害剋溶液灭菌40 min,0.1%HgCl_(2)溶液灭菌5 min;最佳愈伤组织诱导培养基为MS+6-BA 1.5 mg/L+NAA 0.5 mg/L;最佳不定芽增殖培养基为MS+6-BA 2.5 mg/L+NAA 0.2 mg/L;最佳生根培养基为MS+NAA 0.5 mg/L。构建的珠芽黄魔芋无性快繁技术体系在生产中效果明显。In order to solve bottleneck problem of high-quality and large-scale development of Amorphophallus bulbifer industry,taking MS as basic medium and using tubers of A.bulbifer as explants,experiments of explant sterilization,callus induction,adventitious bud proliferation and rooting were carried out,and the best tissue culture and rapid propagation method was selected.Results showed that the best method of explant sterilization was 0.3%hydrocare solution for 40 min and 0.1%HgCl_(2)solution for 5 min.The best medium for callus induc⁃tion was MS+6-BA 1.5 mg/L+NAA 0.5 mg/L.The best medium for adventitious bud proliferation was MS+6-BA 2.5 mg/L+NAA 0.2 mg/L.The best rooting medium was MS+NAA 0.5 mg/L.Asexual rapid prop⁃agation technology system of A.bulbifer was established,and production reaction effect was obvious.
