摘  要:目的 为避免条索形超薄切片样本定位包埋时样本位置发生偏移,设计一种定位包埋模具,以提高样本定向包埋及其超薄切片的效率。方法 在硅胶包埋板凹槽的底部顶端中间位置切割深约1 mm截面为等腰三角形的竖槽或横槽。将完成树脂渗透的条索形样本放入槽中,然后进行包埋、聚合、半薄切片或超薄切片及染色,最后用光学显微镜及透射电子显微镜进行观察。结果 使用常规包埋板包埋的样本约25%出现了位移,使用定位包埋板包埋的样本未出现显著位移,与常规包埋板相比样本位移比例显著下降(P <0. 01)。结论 设计了一种可用于条索形超薄切片样本的定位包埋板,可用于多种组织的定向超薄切片制样。Objective To design a kind of mould for positioning embedding of strip-shaped tissue which to avoid sample shifting during resin polymerization,so as to improve the efficiency of positioning embedding and ultrathin sectioning.Methods Vertical or horizontal slots(1 mm in depth)with isosceles triangle sections were cut at the bottom of silica gel mould along its end.Then the resin infiltrated strip-shaped tissue samples were put into the slots,and followed by embedding,polymerization,semithin or ultrathin sectioning and staining.Finally,sections were observed under optical microscope and transmission electron microscope.Results Displacement appeared in about 25% embedding samples in routine embedding plate while the embedding samples in slots of modified mould didn't shift significantly;compared with the routine embedding plate,there was a significant decline of displacement(P<0.01).Conclusion A kind of positioning embedding plate was designed and could be applied for many kinds of strip-shaped tissue samples.
