摘 要:目的:探究藏药俄色果的降血脂活性。方法:采用体外(医用脂肪乳诱导建立高脂细胞模型)和体内(高脂饲料诱导建立高脂血症小鼠模型) 2种高脂模型,进行俄色果的降血脂活性研究;同时,考察同等剂量的俄色果、俄色叶、山楂对高脂小鼠体重、脏器指数、总胆固醇(total cholesterol, TC)、三酰甘油(triglyceride, TG)、高密度脂蛋白胆固醇(high-density lipoprotein cholesterol, HDL-C)、低密度脂蛋白胆固醇(low density lipoprotein cholesterol, LDL-C)、谷丙转氨酶(alanine aminotransferase, ALT)、谷草转氨酶(glutamic oxaloacetic transaminase, AST)水平的影响,对比评价俄色果的降血脂效果。结果:在医用脂肪乳诱导的高脂细胞模型中,俄色果给药(50,100μg·mL^(-1))显著降低脂变性细胞内TC和TG水平;明显改善脂变性细胞的细胞形态。在高脂饲料诱导的高脂血症小鼠模型中,俄色果给药(0.75,1.5 g·kg^(-1),ig)能减缓高脂小鼠体重增长,显著降低肝脏指数及TC,TG,LDL-C,ALT,AST水平,升高HDL-C水平;同等剂量下,降TC效果比较山楂>俄色果>俄色叶;降TG,LDL-C,ALT效果比较俄色果>山楂>俄色叶;降AST效果比较:俄色果>俄色叶>山楂;升HDL-C效果比较:俄色叶>俄色果>山楂。结论:俄色果具有较好的降血脂活性,能显著改善血脂水平,减轻高脂饮食造成的肥胖症状,同时缓解肝损伤,且在降TG,LDL-C,ALT方面优于俄色叶和山楂。Objective: To investigate the hypolipidemic activity of Tibet medicine "Malus Fructus". Methods: The hypolipidemic activity of Malus Fructus was studied by using two kinds of hyperlipidemia models: in vitro(hyperlipidemia cell model induced by medical fat milk) and in vivo(hyperlipidemia mouse model induced by high fat feed). At the same time, by comparing the effects of same dose of Malus Fructus, Malus Folium, hawthorn on the body weight, viscera index, blood levels of total cholesterol(TC), triglyceride(TG), high-density lipoprotein cholesterol(HDL-C), low density lipoprotein cholesterol(LDL-C), alanine aminotransferase(ALT), and glutamic oxaloacetic transaminase(AST) of high fat mice, the blood lipid-lowering effect of Malus Fructus was evaluated. Results: In the model of hyperlipidemia induced by medical fat milk, the levels of TC and TG in steato-degeneration cells were significantly decreased by the administration of Malus Fructus(50, 100 μg·mL^(-1)), and it obviously improved the morphology of steato-degeneration cells. In the mouse model of hyperlipidemia induced by high-fat diet, the administration of Malus Fructus(0.75, 1.5 g·kg^(-1),ig) slowed down the weight gain of high-fat mice, significantly reduced the liver index and TC, TG, LDL-C, ALT, AST levels, and increased HDL-C levels. Under the same dose, the effects of reducing TC of different materials ranked in the following sequence: hawthorn > Malus Fructus > Malus Folium;the effects of lowering TG, LDL-C and ALT were as follows: Malus Fructus > hawthorn > Malus Folium;AST lowering effect: Malus Fructus > Malus Folium > hawthorn;HDL-C increasing effect: Malus Folium > Malus Fructus > hawthorn. Conclusion: Malus Fructus has good activity of lowering blood lipid, which can significantly decrease blood lipid level, relieve obesity symptoms caused by high-fat diet, and alleviate liver injury. In addition, it is better than Malus Folium and hawthorn in lowering TG, LDL-C and ALT.
