摘 要:目的探讨circ_0081664对非小细胞肺癌细胞生长和转移的影响及机制。方法选取2016年1月至2020年5月期间本院收治的30例非小细胞肺癌患者癌组织及相应的癌旁组织,RT-qPCR检测circ_0081664和miR-1205的表达水平;A549细胞分为si-NC组、si-circ_0081664组、miR-NC组、miR-1205组、si-circ_0081664+anti-miR-NC组、si-circ_0081664+anti-miR-1205组。MTT检测细胞活性;流式细胞术检测细胞凋亡;Transwell小室实验检测迁移和侵袭;Western blot法检测蛋白表达;双荧光素酶报告实验检测circ_0081664和miR-1205的靶向关系。结果肺癌组织中circ_0081664表达水平升高,miR-1205表达水平降低(P<0.05)。转染circ_0081664抑制表达载体或miR-1205过表达载体后,A549细胞活性降低,Ki-67、Bcl2、MMP-2、MMP-9表达水平降低,P21、Bax表达水平升高,细胞凋亡率升高,迁移侵袭细胞数减少(P<0.05)。同时抑制circ_0081664和miR-1205表达后,细胞活性升高,细胞凋亡率降低,迁移侵袭细胞数增加(P<0.05)。结论抑制circ_0081664表达可能通过上调miR-1205,抑制非小细胞肺癌细胞的生长和转移。Objective To explore the effect and mechanism of circ_0081664 on the growth and metastasis of non-small cell lung cancer cells. Methods The cancer tissues and corresponding paracancerous tissues of 30 patients with non-small cell lung cancer admitted to our hospital from January 2016 to May 2020 were selected. A549 cells were divided into the si-NC group, the si-circ_0081664 group, the miR-NC group, the miR-1205 group, the si-circ_0081664+anti-miR-NC group, and the si-circ_0081664+anti-miR-1205 group. It used RT-qPCR to detect the expression levels of circ_0081664 and miR-1205, MTT to detect cell viability, flow cytometry to detect cell apoptosis, Transwell chamber experiment to detect the cell number of migrated and invaded, Western blot method to detect protein expression, and dual luciferase report experiment to detect the targeting relationship between circ_0081664 and miR-1205. Results The expression level of circ_0081664 was increased in lung cancer tissues, and the expression level of miR-1205 was decreased(P<0.05). After transfection with circ_0081664 suppression expression vector or miR-1205 overexpression vector, the cell viability was decreased, the expressions of Ki-67, Bcl2, MMP-2, MMP-9 were decreased, the expressions of P21 and Bax were increased, and the apoptosis rate was increased, and the number of migrating and migrating cells was decreased(P<0.05). After simultaneously inhibiting the expression of circ_0081664 and miR-1205, the cell viability was increased, the cell apoptosis rate was decreased, and the number of migrating cells was increased(P<0.05). Conclusion Inhibiting the expression of circ_0081664 may inhibit the growth and metastasis of non-small cell lung cancer cells by up-regulating miR-1205.
