摘 要:目的探究人胎盘源间充质干细胞(human placenta derived mesenchymal stromal cells,hPMSCs)对人急性淋巴白血病细胞(Jurkat T)表面CD4、CD28及CD175表达的影响及机制。方法将hPMSCs与Jurkat T细胞共培养,采用FCM检测共培养前后Jurkat T细胞表面CD4、CD28、CD175的表达水平;Western blot检测Cosmc蛋白表达水平,荧光法检测T-合酶(T-synthase)活性,细胞O-糖组扩增技术(Cellular O-glycome Reporter/Amplifition,CORA)检测O-糖组变化。结果hPMSCs O-糖组结构较复杂,表达核心1来源的O-糖链结构(m/z955.4、1129.5、1316.6)及核心2来源的O-糖链结构(m/z1217.6、1648.7、1782.8、1939.9、2058.0、2563.2);Jurkat T细胞不表达核心1及核心2来源的O-糖链。与hPMSCs共培养后,Jurkat T细胞表面CD4表达增高,CD28、CD175表达降低;Cosmc蛋白表达增多,T-合酶活性增高,表达核心1来源的O-聚糖结构(m/z955.4,1316.6)。结论hPMSCs可通过上调Cosmc蛋白、T-合酶活性,改变Jurkat T细胞O-糖基化状态影响其表面CD4、CD28及CD175表达。Objective To explore the effect of human placenta derived mesenchymal stromal cells(hPMSCs)on CD4,CD28 and CD175 expression in human acute lymphatic leukemia cells(Jurkat T)and its mechanism.Methods HPMSCs were co-cultured with Jurkat T cells,and FCM was used to detect the expression levels of CD4,CD28 and CD175 on Jurkat T cells before and after the co-culture.Western blot was used to detect Cosmc protein,fluorescence method was used to detect T-synthase activity,and Cellular O-glycome Reporter/Amplifition(CORA)was used to detect the changes of O-glycans.Results The structure of hPMSCs O-glycan is complex,including the core 1 derived O-glycan structures(m/z:955.4,1129.5,1316.6)and the core 2 derived O-glycan structures(m/z:1217.6,1648.7,1782.8,1939.9,2058.0,2563.2).Jurkat T cells do not express core 1 and 2 derived O-glycan structures.After co-cultivation with hPMSCs,its CD4 increased,whereas,CD28 and CD175 decreased.Its Cosmc protein and T-synthase activity increased,with the expression of the core 1 derived O-glycan structures(m/z:955.4,1316.6).Conclusion HPMSCs could affect the expression of CD4,CD28 and CD175 on the surface of Jurkat T cells by changing the status of O-glycosylation.
