摘 要:目的探讨良性脑膜瘤及柔脑膜原代细胞的培养及脂多糖(LPS)对其增殖的影响。方法通过酶消化法培养良性脑膜瘤及柔脑膜原代细胞,采用免疫荧光组化方法鉴定脑膜瘤细胞。用LPS处理原代细胞后,采用CCK法检验细胞增殖活性,Real-timePCR对常见的炎症因子如白细胞介素1β(IL-1β)、白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α)及核因子κB(NF-κB)mRNA表达水平进行检测。结果成功培养良性脑膜瘤及柔脑膜细胞原代细胞,免疫荧光鉴定细胞均表达上皮膜抗原(EMA)和波形蛋白(VIM)。LPS处理细胞后,细胞的增殖活性随LPS浓度增加和时间延长而增加,炎症因子IL-1β、IL-6、TNF-α和NF-κB mRNA表达水平也明显升高。结论良性脑膜瘤与柔脑膜原代细胞的培养成功为良性脑膜瘤的基础研究打下了基础。LPS处理细胞后,细胞增殖明显增加,炎性因子和NF-κB表达上调。良性脑膜瘤的生长与炎症有密切关系。Objective To investigate the culture of benign meningioma and leptomeningeal primary cells and the effects of lipopolysaccharide(LPS)on their proliferation.Method Primary benign meningioma and leptomeningeal cells were cultured by chemical digestion.Immunofluorescence was used to identify the meningioma cells.Cell proliferation was tested by CCK-8.The mRNA expression of inflammatory factors such as IL-1β,IL-6,TNF-αand NF-κB was tested by real-time PCR.Results The primary cells of benign meningioma and leptomeningeal cells were successfully cultured.Immunofluorescence identification showed that the cells expressed epithelioma antigen(EMA)and vimentin(VIM).After treatment with LPS,the cell proliferation was up-regulated with time and concentration,and the expression of inflammatory factors,such as IL-1β,TNF-α,IL-6 and NF-κB were increased.Conclusions The success of primary benign meningioma and leptomeningeal cell culture would lay the foundation for further basic research of benign meningioma.After LPS treatment,cell proliferation increases significantly,inflammatory factors and NF-κB expression is up-regulated.And the growth of benign meningiomas has a close relationship with inflammation.
