摘  要:以精细化学品开发为目标,融合“绿色化学”理念且符合课堂教学条件要求的综合实验案例,在化学类高等人才培养体系中仍然匮乏。本文设计并开发了一个新型、绿色合成香兰素及香兰素乙酸酯的大学化学综合实验。选取粗丁香油为原料,酸碱萃取法提纯得到丁香酚后,在空气参与下,铁催化/氧化断裂烯丙基制备得到香兰素,酚羟基乙酰化后制备得到香兰素乙酸酯。最后,采用红外、核磁波谱分析、三氯化铁特异性识别、香料评价等手段对产物结构进行表征和评价。本实验不仅具有绿色环保、操作安全等优点,而且可以让学生在学习、掌握有机合成、波谱分析等理论知识的同时,通过“多手段结构验证”、“闻香识化学”等环节提高了实验的科学性和趣味性,培养了学生的创造性和发散思维。Comprehensive experimental teaching cases that target fine chemical preparation,integrate the concept of"green chemistry"and meet the requirements of classroom teaching conditions,are still lacking in the training system for advanced chemistry students.Herein,a novel comprehensive chemistry experiment for the synthesis of vanillin and acetyl vanillin was designed and developed.In this experiment,crude clove oil was chosen as the raw material.After the eugenol was purified by the classical acid-base extraction method,the allyl group of eugenol was oxidized and cleaved by the iron catalyst to generate vanillin in the presence of air.Next,acetyl vanillin was obtained in good yield via acetylation of the phenolic hydroxyl group.The products were characterized and evaluated by IR/1H NMR analysis,specific identification of ferric chloride,and perfume evaluation.This experiment not only has the advantages of environmentally friendly and operational safety but also increased the scientificity and interest of the experiment through the designing parts of the"multi-method structural elucidation"and"scent of the chemistry".
