摘 要:目的:探讨锌指蛋白GATA6(GATA6)小分子干扰RNA(siRNA)对肝癌HCC-9204细胞自噬和周期的影响及机制。方法:肝癌HCC-9204细胞中转染GATA6 siRNA、siRNA control,定量即时聚合酶链锁反应(qRT-PCR)方法检测GATA6 mRNA表达;Western blot方法检测GATA6、卷曲螺旋状Myosin样Bcl-2相互作用蛋白(beclin1)、自噬相关基因3(ATG3)、磷酸化信号转导与转录因子3(p-STAT3)蛋白表达;流式细胞术检测细胞周期。STAT3信号激活剂处理转染GATA6 siRNA后的肝癌HCC-9204细胞,用上述方法检测细胞周期和beclin1、ATG3、p-STAT3蛋白表达。结果:与转染siRNA control后的肝癌HCC-9204细胞比较,转染GATA6 siRNA可以降低细胞中GATA6 mRNA和蛋白表达量,提高细胞G0/G1期比例,减少细胞中beclin1、ATG3、p-STAT3蛋白表达。STAT3信号激活剂可以降低转染GATA6 siRNA后的肝癌HCC-9204细胞G0/G1期比例,提高细胞中beclin1、ATG3、p-STAT3蛋白表达水平。结论:GATA6 siRNA抑制肝癌HCC-9204细胞自噬并阻滞细胞周期,作用机制与降低STAT3信号通路激活水平有关。Objective:To study the effect and mechanism of GATA Binding Protein 6(GATA6)smallinterfering RNA(siRNA)on autophagy and cycle of hepatoma HCC-9204 cells.Methods:Hepatoma HCC-9204 cells were transfected with GATA6 siRNA and siRNA control,and Quantitative real time polymerase chain reaction(qRT-PCR)was used to detect GATA6 mRNA expression.Western blot was used to detect GATA6,Coiled-coil,Myosin-likeBcl-2-Interacting Protein(beclin1),autophagy related gene 3(ATG3),and phosphorylated signal transducers and activators of transcription 3(p-STAT3)protein expressions.Flow cytometry was used to detect cell cycle.Hepatoma HCC-9204 cells transfected with GATA6 siRNA were treated with STAT3 signal activator,and the cell cycle and beclin1,ATG3,p-STAT3 protein expression were detected.Results:Compared with hepatoma HCC-9204 cells transfected with siRNA control,transfection of GATA6 siRNA could reduce the expression of GATA6 mRNA and protein in cells,increase the ratio of G0/G1 phase,and reduce the expression of beclin1,ATG3,and p-STAT3 proteins in cells.STAT3 signal activator could reduce the G0/G1 phase ratio of hepatoma HCC-9204 cells transfected with GATA6 siRNA,and increase the expression of beclin1,ATG3,and p-STAT3 proteins in the cells.Conclusion:GATA6 siRNA inhibits autophagy of HCC-9204 cells and blocks cell cycle,the mechanism of action is related to reducing the activation level of STAT3 signaling pathway.
