基于数据库探讨NEDD8在头颈鳞癌中的表达及其预后价值

摘　　要：目的通过数据库探讨神经前体细胞表达下调因子8(NEDD8)在头颈鳞癌中的表达及其预后价值。方法利用Oncomine和UALCAN数据库分析NEDD8在头颈鳞癌组织和正常组织中的表达差异;运用GEPIA数据库和Kaplan-Meier Plotter在线分析NEDD8表达与头颈鳞癌患者生存率的关系;采用cBioPortal数据库分析头颈鳞癌患者中NEDD8基因改变情况。进一步通过LinkedOmics数据库寻找NEDD8共表达基因,并进行基因本体论(GO)和京都基因与基因组百科全书(KEGG)通路富集分析。结果NEDD8 mRNA在头颈鳞癌中表达水平显著高于正常组织,且与患者临床肿瘤分期、病理肿瘤分级、淋巴转移分期等显著相关(均P<0.05)。生存曲线分析显示,NEDD8高表达组头颈鳞癌患者总生存率显著低于低表达组(P<0.05)。NEDD8突变与HNSCC患者生存率无明显关系。NEDD8基因可能参与翻译延伸和蛋白折叠等生物学过程,并在核糖体、蛋白运输和嘧啶代谢通路等方面富集。结论NEDD8基因在头颈鳞癌患者中高表达且提示患者预后较差。NEDD8基因可能是潜在的头颈鳞癌生物标志物和治疗新靶点。Objective To explore the expression and prognostic value of NEDD8 gene in head and neck squamous cell carcinoma(HNSCC)by data mining.Methods The expression of the NEDD8 in HNSCC and normal tissues were analyzed using the Oncomine and UALCAN databases.And its correlation with the prognosis of patients were further investigated via GEPIA and Kaplan-Meier.The variations of NEDD8 in HNSCC were analyzed by cBioPortal database.Furthermore,the NEDD8 co-expression genes were screened through the LinkedOmics database,and GO enrichment and KEGG pathway analysis were performed.Results NEDD8 mRNA was highly expressed in HNSCC and significantly associated with clinical cancer stage,tumor grade and nodal metastasis status in patients(P<0.05).High expression of NEDD8 indicated a poor prognosis(P<0.05).There was no significant correlation of NEDD8 mutation with the survival rate of HNSCC patients.The NEDD8 gene may be involved in biological processes including translational elongation and protein folding,and enriched in ribosome,protein export and pyrimidine metabolism.Conclusion NEDD8 gene is highly expressed in HNSCC patients and indicates a poor prognosis,suggesting that the gene may serve as a potential biomarker and new therapeutic target of HNSCC.
