摘  要:目的明确血管性认知损伤大鼠脑内SIRT1表达及氧化应激指标SOD及MDA的变化情况。方法2-VO法建立慢性脑缺血大鼠模型,随机分为假手术组和慢性脑缺血组,于术后6周、9周、12周,利用Morris水迷宫试验进行认知评估,实时定量PCR、免疫组化及Western blot检测大鼠脑内SIRT1表达,同时进行SOD、MDA水平测定。结果2-VO术后6周、9周和12周,慢性脑缺血大鼠SIRT1蛋白表达明显下降,以缺血6周时最明显;慢性脑缺血时大鼠脑内SOD降低而MDA升高,显著差异仅发生在脑缺血6周。结论慢性脑缺血时SIRT1表达活性持续受到抑制,早期的氧化应激反应可能参与了大鼠认知损伤的的发生发展过程,有望成为血管性认知损伤的干预新靶点。Objective To determine the changes of SIRT1expression and SOD and MDA levels of oxidative stress index in rats brain with vascular cognitive impairment.Methods Healthy male Wistar rats were randomly divided into sham operation(SH)groups and chronic cerebral ischemia(CCI)groups.After chronic cerebral ischemia rat models induced by 2-VO method were established,all rats received behavioral testing with Morris water maze(at different survival times of 6weeks,9weeks,12weeks after 2-VO).After that the expression of SIRT1proteins in frontal cortex and hippocampus were detected by real time PCR,immunohistochemical staining and Western blot assay.Meanwhile SOD and MDA in Hippocampus and frontal lobe were determined according to the specification of the kits.Results The cognitive impairment of CCI rats changed dynamically with the prolongation of ischemia time at 6,9and 12weeks after 2-VO operation.There was no significant decrease in SIRT1RNA level in rats with chronic cerebral ischemia,but SIRT1 protein levels of 2-VO rats were significantly decreased,mostly in 6-weeks CCI group.The levels of SOD decreased,while the MDA levels increased after CCI,and the significant differences occurred only at 6weeks of cerebral ischemia.Conclusion Early reaction of oxidative stress and continuously restrained SIRT1activity may be involved in the occurrence and development of cognitive impairment induced by CCI,who were expected to be a new intervention target for vascular cognitive impairment.
