摘  要:试验研究产酸性纤维素酶菌株的筛选和纤维素酶的酶学性质。试验以羧甲基纤维素钠为唯一碳源,从宜宾竹鼠粪便中采用刚果红染色法初筛和DNS法测定纤维素酶活进行复筛,筛选得到1株产酸性纤维素酶的菌株G-1,使用16S rDNA序列分析,鉴定为干燥杆菌属(Siccibacter colletis),对菌株G-1所产纤维素酶的酶学性质初步研究。结果显示,纤维素酶最适反应温度为40℃,最适pH值为4.0,在pH值为4~6的范围内保持1 h,相对酶活力在85%以上,对酸性环境的抗受性强;在30~50℃温度范围中静置1 h,相对酶活力在90%以上;Mn^(2+)对该纤维素酶活有促进作用,Fe^(3+)对其抑制效果明显。研究表明,试验所得菌株G-1的纤维素酶在酸性环境下具备良好的活性,能够丰富饲料中纤维素酶的种质库资源,作为饲料中酶添加剂具有的巨大潜力。The experiment was to study screening of acid cellulase strain degradation resistance in bamboo rats and study on enzymatic properties.Sodium carboxymethyl cellulose was used as the only carbon source,and cellulase activity was determined by the primary screening method of the Congo red staining and the re-screening method of the DNS method from the bamboo manure of Yibin.A strain of acid cellulase-producing strain G-1,16S rDNA sequence analysis,was identified as Siccibacter Colletis.The enzymatic properties of cellulase produced by strain G-1 were studied.The results showed that the optimal reaction temperature of cellulase was 40℃,the optimal pH value was 4.0,and the relative enzyme activity was more than 85% in range of pH value 4~6 for 1 h,and the resistance to acidic environment was strong.The relative enzyme activity was more than 90% after standing for 1 h in temperature range of 30~50℃.Mn^(2+) promoted the cellulase activity,while Fe^(3+) significantly inhibited the cellulase activity.The experiment indicates the cellulase of strain G-1 obtained in study has good activity under acidic environment,which enriches cellulase seed bank resources in feed and has great potential as enzyme additive in feed.
