摘  要:为了解我国J亚群禽白血病病毒(ALV)的流行特点和进化趋势,本试验从河北某鸡场疑似禽白血病的临床发病鸡的病料组织中,分离得到2株ALV-J毒株,分别命名为CAU2019和CAU2020。通过聚合酶链式反应(PCR)技术分段扩增分离株的全病毒基因组,连接pEASY-Blunt Zero载体后进行序列测定,并对获得的基因组序列与ALV各亚群毒株序列进行序列比对及遗传进化分析。结果表明,2个分离株与ALV各亚群毒株的gag和pol段基因相对保守,而LTR、gp85及gp37段基因变异较大。此外,CAU2019在3′UTR处有2段长度为30 bp和11 bp的不连续的碱基缺失,CAU2020在3′UTR处有1段长度为29 bp的碱基缺失。由gp85和LTR基因构建的遗传进化树可知,2个分离株在遗传关系上与J亚群毒株JS16JH9亲缘关系最近。结果表明本试验分离的2株病毒均为J亚群禽白血病病毒毒株,并且为国内ALV-J流行株的突变株。This study aimed to understand the epidemiological characteristics and evolutionary trends of the subgroup J avian leukemia virus(ALV) in China. Two strains of ALV-J,named CAU2019 and CAU2020, were isolated from the diseased tissues of clinically ill chickens suspected of avian leukemia in a chicken farm in Hebei. The whole genomes of the isolates were amplified by the polymerase chain reaction(PCR) technique in segments, and the sequence determination was performed after ligating the PCR products into pEASY-Blunt Zero vector. The resulting genomic sequences were compared with the sequences of ALV subgroups and analyzed for genetic evolution. The results showed that the gag and pol segments were relatively conserved between these 2 isolates and the ALV subgroups. In contrast, the genes of LTR,gp85 and gp37 segments were more variable. Additionally, CAU2019 contained 2 discontinuous base deletions of 30 bp and 11 bp in 3′UTR, while CAU2020 contained a deletion of 29 bp in 3′UTR. The phylogenetic tree analysis based on gp85 and LTR genes revealed that the 2 isolates have the closest genetic relationship with subgroup J strain JS16 JH9. These findings suggest that the 2 isolates belong to subgroup J avian leukemia virus and represent mutants of the prevalent domestic ALV-J strain.
