摘 要:目的提取动静脉畸形(Arteriovenous malformation,AVM)内皮细胞并与人脐静脉内皮细胞(Human umbilical vein endothelial cells,hUVEC)对比,观察其基本功能特征。方法从颅外AVM病灶组织中分离并提取内皮细胞,通过CCK⁃8法评估AVM内皮细胞增殖速度、小管形成实验评估其血管生成能力以及划痕实验评估其迁移能力。所有实验均以hUVEC作为对照。结果在增殖能力上AVM内皮细胞与hUVEC相近,在迁移能力上则较弱。在成管能力方面,AVM内皮细胞形成的管较hUVEC形成的管长且围成的管腔面积也较大。结论颅外AVM血管内皮细胞较弱的迁移能力及异常的成管能力可能与AVM异常的血管形成有关。Objective To extract endothelial cells of arteriovenous malformation(AVM)and compare with human umbilical vein endothelial cells(hUVEC)to observe their basic functional characteristics.Methods Endothelial cells were isolated and extracted from the lesion tissues of extracranial arteriovenous malformations.The proliferation rate of AVM endothelial cells was evaluated by CCK⁃8.The ability of angiogenesis was evaluated by tubule formation assay.The ability of migration was evaluated by scratch assay.hUVEC was used as control in all experiments.Results The endothelial cells of AVM were similar to those of hUVEC in proliferation ability,but weak in migration ability.In terms of tubular capacity,the tubes formed were longer than those formed by hUVEC,and the area which enclosed by tubules was larger.Conclusion The weak migration and abnormal tube formation of vascular endothelial cells in extracranial arteriovenous malformations may be related to the abnormal angiogenesis of arteriovenous malformations.
