摘 要:研究通过同源重组的方法,敲除产植酸酶毕赤酵母菌株的醇氧化酶aox1基因,以改变菌株利用甲醇的速度,降低甲醇用量。试验得到一株甲醇利用慢型毕赤酵母,能降低发酵成本。在实际发酵生产中,aox1基因缺失的菌株发酵能力与原有菌株接近,生产能耗大幅降低,甲醇用量降低,最终单罐发酵成本降低约20%~24%。In this study,the alcohol oxidase aox1 gene of phytase-producing Pichia pastoris strain was knocked out by homologous recombination method,with the purpose to change the rate of methanol utilization and reduce the methanol consumption.Finally,a methanol utilization slow Pichia pastoris strain was obtained and the fermentation cost was reduced.In the actual fermentation production,the fermentation capacity of the recombinant strain was close to the original strain,but the production energy consumption was greatly reduced,the methanol consumption was reduced,and the final cost of single-tank fermentation was reduced by about 20%-24%.
