摘  要:目的:研究常氧及低氧条件下SHG-44胶质瘤细胞中活性氧(ROS)及低氧诱导因子-1α(HIF-1α)的表达及意义。方法:在常氧条件及低氧条件下培养SHG-44胶质瘤细胞,分为常氧对照组、常氧NAC干预组、低氧对照组、低NAC氧干预组四组。常氧及低氧干预组加入N-乙酰半胱氨酸(NAC),48 h后检测ROS的表达,RT-PCR检测HIF-1αmRNA表达。结果:低氧对照组ROS表达水平明显高于常氧对照组(P<0.05),常氧NAC干预组及低氧NAC干预组给予抗氧化剂NAC之后ROS表达水平均明显下降(均P<0.05);低氧对照组HIF-1αmRNA表达高于常氧对照组、常氧NAC干预组及低氧NAC干预组(均P<0.05)。结论:低氧条件下ROS及HIF-1αmRNA表达水平明显高于常氧条件,但是应用ROS干预剂NAC之后低氧条件下SHG-44胶质瘤细胞中HIF-1αmRNA表达下降,提示ROS在促进HIF-1αmRNA表达中起重要作用。Objective:To study the expression and significance of reactive oxygen species(ROS)and hypoxia inducible factor-1α(HIF-1α)in SHG-44 glioma cells under normoxia and hypoxia.Methods:SHG-44 glioma cells were cultured under normoxia and hypoxia conditions and divided into normoxia control group,normoxia intervention group,hypoxia control group,and hypoxia intervention group.The normoxia and hypoxia intervention groups were added with N-acetylcysteine(NAC),and the expression of ROS was detected 48 hours later,and the expression of HIF-1αmRNA was detected by RT-PCR.Results:The expression level of ROS in the hypoxic control group was significantly higher than that of the normoxic control group(all P<0.05).The expression of ROS in the normoxia intervention group and the hypoxia intervention group was significantly reduced after the antioxidant NAC was given(all P<0.05).The expression of HIF-1αmRNA in the hypoxia control group was higher than that of the normoxia control group,the normoxia intervention group and the hypoxia intervention group(all P<0.05).Conclusion:The expression levels of ROS and HIF-1αmRNA under hypoxic conditions were significantly higher than those under normoxia conditions.However,the expression of HIF-1αmRNA in SHG-44 glioma cells decreased under hypoxic conditions after the application of NAC,an intervention agent of ROS.ROS play an important role in promoting the expression of HIF-1αmRNA.
