摘　　要：目的探讨三类主要细胞骨架相关蛋白在小鼠神经系统中表达和分布。方法以1.5个月龄C57BL/6J野生型小鼠为材料,使用蛋白印迹法检测微管、微丝和神经丝相关蛋白在小鼠眼、脑、脊髓、坐骨神经和肌肉中表达情况。以6个月龄小鼠脊髓切片为材料,使用免疫荧光和和激光共聚焦显微镜技术检测细胞骨架蛋白的表达和定位情况。结果微管、微丝和神经丝相关蛋白在小鼠神经系统中表达广泛,且具有差异性。与脊髓相比,坐骨神经中大部分细胞骨架蛋白含量相对下降(P<0.05)。结论微管、微丝和神经丝相关蛋白是神经元主要细胞骨架蛋白,在神经系统广泛表达,但具有组织和亚组织差异化表达和分布,这可能与神经元结构和功能特异性有关。Objective To investigate the expression and distribution of cytoskeleton-associated proteins(CSAP)in the nervous system in mice.Methods The tubulin,actin and neurofilament related proteins distribution in eye,brain,spinal cord,sciatic nerve and muscle,and the expression of CSAP in spinal cord and sciatic nerve of 1.5 months wild-type mouse were examined by western blot.The distribution of CSAP in spinal cord of 6 months mouse were examined by immunofluorescence and laser confocal scanning microscopy.Results CSAP had different expression levels and distribution in mouse nervous system.Compared to spinal cord,the sciatic nerve showed decreased level of most CSAP(P<0.05),and neurofilament showed comparable level.Conclusion tubulin,actin and neurofilamen are major CSAP,and are widely and differently distributed in the nervous system of mice,which is related to the structural and functional specificity of neurons.
