摘  要:目的探讨鼠双微体基因2(MDM2)和高迁移率族蛋白B1(HMGB1)在骨软骨瘤组织和骨肉瘤组织中的表达及其临床意义。方法收集2016年1月至2020年12月辽宁省人民医院和中国医科大学附属盛京医院资料病理学确诊的27例骨软骨瘤组织和80例骨肉瘤组织标本,应用免疫组织化学法检测上述组织MDM2蛋白和HMGB1蛋白的表达水平,采用χ^(2)检验进行统计学分析。结果骨肉瘤组织中MDM2表达率为68.75%(55/80)、明显高于骨软骨瘤组织MDM2表达率[7.41%(2/27)],两者差异有统计学意义(χ^(2)=30.515,P<0.01),MDM2表达水平与骨肉瘤恩内金分期(Eneeking分期)、肺转移明显相关(χ^(2)=5.1239、4.4649,P<0.05),差异有统计学意义。骨肉瘤组织中HMGB1表达率为56.25%(45/80)、明显高于骨软骨瘤组织中HMGB1表达率[14.82(4/27)],两者差异有统计学意义(χ^(2)=13.962,P<0.01)。HMGB1表达水平与骨肉瘤Eneeking分期、肺转移、软组织浸润明显相关(χ^(2)=5.261、4.127、4.898,P<0.05),差异有统计学意义。结论 MDM2蛋白和HMGB1蛋白表达与骨肉瘤转移、侵袭明显相关,检测MDM2和HMGB1有助于诊断骨肉瘤患者病情进展。Objective To evaluate the expression of murine double minute 2(MDM2)and high mobility group box 1(HMGB1)in osteosarcoma tissue and osteochondroma tissue and their clinical significance.Methods 27 cases of osteochondroma tissues and 80 cases of osteosarcoma tissues of patients pathologically confirmed from january 2016 to december 2020 on Liaoning Provincial People′s Hospital and Shengjing Hospital of China Medical University were collected,the expression of MDM2 protein and HMGB1 protein in osteochondroma tissues and osteosarcoma tissues were detected by using immunohistochemical method,Chi-square test was used for statistical analysis.Results The positive rate of MDM2 protein expression in osteosarcoma tissues 68.75%(55/80)was significantly higher than that in osteochondroma tissues 7.41%(2/27),the difference was statistically significant(χ^(2)=30.515,P<0.01).The expressions of MDM2 were related osteosarcoma to Eneeking stage and lung metastasis(Respectively,χ^(2)=5.124,4.465,P<0.05),the difference was statistically significant.the positive rate of HMGB1 protein expression in osteosarcoma tissues 56.25%(45/80)was significantly higher than that in osteochondroma tissues 14.82(4/27),the difference was statistically significant(χ^(2)=13.962,P<0.01).the expressions of HMGB1 were related osteosarcoma to Eneeking stage,lung metastasis and soft tissue involvement(Respectively,χ^(2)=5.261,4.127,4.898,P<0.05),the difference was statistically significant.Conclusion The expression of MDM2 protein and HMGB1 protein were significantly correlated with the metastasis and invasion of osteosarcoma,detection of MDM2 and HMGB1 was helpful for the diagnosis of the disease progression of osteosarcoma patients.
